Isozymes of Alkaline Phosphatases in Developing Embryos of the Hairy Hyalomma Tick

Abstract/ Executive Summary:

Alkaline phosphatase isozymes were recorded apparently for the first time in developing embryos of Hyalomma rufipes. Electrophoresis was carried out in a disc gel apparatus using acrylamide and bis-acorylamide as gels. The staining agents used were P-naphthyl phosphate and Fast blue B-B salt. One type of band was recorded during earlier stages of development and two bands became apparent at later stages of development.

Objectives:

To investigation of isozyme of alkaline phosphates during embryonic development in Hyalomma rufipes.

Methodology:

Eggs were obtained from ovipositing Haylomma rupfies which had been incubated at 26±1⁰C; 85 r. h. Eggs at different stages of development such as cleavage, blastoderm formation, gastrulation segmentation, germ band contraction and pre-hatch was determined with the held of stereo-descting microscope. The above mentioned structural changes were recorded apparently within 1, 7,14, 21, 28 and 33 days of oviposition respectively. Samples from these different developmental stages were homogenised in a ground glass homogenizer using 2.0 mg eggs per ml of distilled water and crude homogenate was used in each occasion. Vertical disc gels were prepared by the methods. Monomer containing 20% acrylamide (B DH) and 08% bis-acrylamide was used; buffers such as separation, staking and electrode were prepared at pH 8.9, 6.9 and 8.3, respectively. Homogenate was stacked in the stacking gell and placed in the tubes to occupy 6 cm linear space of the tubas. Electrophoresis was carried out in a disc gel apparatus containing typically 8 cm tubes of 6 mm internal diameter. The initial voltage was 300 V and the current 6 mA per tube supplied by a 'Vokam' power pack. Electrophoresis was run approximately for two hours till the marker had crossed the gel. The gels were treated for 1 hour at room temperature (18.24^oC) in a staining solution containing B-naphthyl phosphate and Fast blue BB salt at pH 9.7.
