Study on Floral Biology of Jackfruit Germplasm

Abstract/ Executive Summary:

An experiment on floral biology in jackfruit germplasm was carried out at the orchard of Jackfruit Research Project, Department of Horticulture, Bangladesh Agricultural University, Mymensingh during May 2000 to July 2003. The flowering of jack fruit started from the first week of November and continued up to the last week of May. Emergence of anthers on the surface of male spike (anthesis) occurred during day between 10:00 to 18:00h. Maximum anthesis (39.49%) was observed between 14:00 to 16:00h. Dehiscence started at 17:00h and attained the peak (70.29%) from 18:00 to 19:00h and ended at 20:00h on the day of its emergence. The availability of pollen grain on male spike lasted from 4.6 to 6.6 days. The average diameter of pollen grain was 17.34 microns. Germ plasm had high pollen viability ranging from 84.22 to 99.21%. Pollen grains retained their viability for 24 hours when stored at room temperature. Maximum pollen germination was obtained in 10% sucrose solution. Protrusion of stigma started 2 days before and continued up to 10.2 days after opening of spathes. The duration of receptivity of stigmas of a female spike was 26.96 days.

Objectives:

To study on floral biology of Jackfruit germplasm.

Methodology:

The experiment was conducted on selected 12 to 14 years old jackfruit germplasm mainly in the orchard and field laboratory of Jackfruit Research Project, Department of Horticulture, Bangladesh Agricultural University (BAU), Mymensingh during May 2000 to July 2003. Nine germplasm (Nos. J-07, J-11, J-14, J-19/2, J-20, J-30, J-22, J-31/1, J-31/2 and J-51) were selected for the study. The plants were 12 to 13 years of age, healthy and they received similar cultural treatments. The soil of the orchard was silty loam under old Brahmaputra flood plain, fertile, well drained and slightly acidic with pH varying from 5.5 to 6.8. The pollen grains collected from spikes of nine selected jackfruit germplasm were studied for pollen viability and germination test. Pollen grains were collected just after dehiscence from anther and stored at room temperature. The viability was studied under a compound microscope using 0.5% acetocarmine as stain at two hours interval. Three slides at each interval for each germplasm were used to take observations. Plump and deeply stained pollen grains were considered as viable and shriveled, unstained or lightly stained pollen grains were recorded as non-viable. Three male spikes from each germplasm were taken to study the pollen viability. The viability percentage was calculated by counting the viable and non-viable pollen grains in random field covering 300 pollen grains. Pollen grains were collected from nine jackfruit germplasm soon after the dehiscence of anthers. As far as possible pollen from a single inflorescence and when not sufficient, pollen from another inflorescence of same plant was used. The cultural media were prepared by dissolving sucrose with distilled water. Concentrations of aqueous sucrose solutions used for germination were 0.0, 2.5, 5, 10, 15 and 20 per cent. The hanging drop method was utilized for culturing the pollen grains. Four slides under each sucrose solution were used to take observations. The progress of germination was studied at 12 hours interval but final count was taken after 48 hours of planting the pollen. The germination percentage was calculated by counting the germinated and ungerminated pollen grains in random field covering 300 grains. Pollen grains from freshly dehisced anther were collected. Then pollen grains were placed in glycerin jelly media in slides covered by zero power cover slips. The diameters of pollen grains were measured by stage and ocular micrometer using 45x objective and 10x eye-piece. Period between receptivity of first stigma to the receptivity of the last stigma on the female spike was considered as duration of receptivity of female spike. Start of receptivity of a stigma was considered as its stickiness and its end by withering. Receptivity of female spike was recorded by day to day observation of 5 female spikes at the time of protrusion of stigma to withering by using magnifying glass.

The information obtained from the study on jackfruit spikes, pollen biology and receptivity of stigma will be useful for the improvement of the crop through hybridization work.
