Embryonic and Larval Development of Indigenous Sarpunti (puntius Sarana, Hamilton)

Abstract/ Executive Summary:

Early development stages of endangered Puntius sarana were described to know the life history of the fish. First cleavage was recorded within 35 minutes after fertilization and the embryonic rudiments of developing eggs appeared at 9 hrs and 40 minutes at 27.0 - 27.5^oC. The yolk sac was completely absorbed at 66 hrs of the development on attainment of 6.0 mm total length.

Objectives:

To highlight some aspects of the early life history of P. sarana.

Methodology:

The experiment was conducted in the hatchery of field laboratory complex and in the water quality laboratory of the Department of Fisheries Management, Bangladesh Agricultural University (BAU), Mymensingh. Sarpunti (P. sarana) were induced to spawn through hypophysation by PG extract at the dose 6.5 mg PG/kg body weight of female. The fertilized eggs were washed with Fertilization solution (4g NaCl and 3g Urea L^-1water) and Tannin solution (0.5g L^-1water) for several time; to remove stickiness and finally transferred into hatching jars with continuous water circulation. The developing stages of sarpunti were observed at every 10 minutes interval till the completion of morula and then every 1hr interval up to hatching. The eggs were preserved into 70% ethanol solution for further study. Early developmental stages were studied under a Stereomicroscope (Olympus, SZH 10 Research Stereo). Development stages were studied continuously until the embryo started twisting movement, which took about eighteen to twenty hours. For clear observation, individuals were temporarily stained with methelene blue. The specimens were measured by placing them over a petridish having 1.0 mm graph paper at the bottom. Embryonic and larval stages were examined at 26.6-27.5 ^oC temperature. Five specimens were used to describe each stage.
