Six carbon and six nitrogenous sources and seven pH levels were tested for their effect on growth and sclerotia formation of Rhizoctonia oryzae-sativae under in-vitro condition. Out of six carbon sources tested, dextrose and sucrose provided the best results for growth and sclerotia formation followed by galactose and fructose. Among six different nitrogen sources, peptone was found best for vegetative growth of Rhizoctonia oryzaesativae followed by yeast and NaNO3. Studies on pH revealed that the tested pathogen grew maximum growth at 6.0-9.0 on Potato dextrose agar (PDA) medium. The results of the present investigation indicated that nutritional condition like carbon and nitrogen sources as well as physical factors such as pH greatly affected the growth and production of sclerotia by Rhizoctonia oryzae-sativae.

The study is aimed to find out useful information about the effect of different carbon and nitrogen sources and also different pH level on mecelial growth and sclerotia formation of R. oryzaesativae under in vitro condition.

This experiment was conducted in the Plant Pathology Division, BRRI, Gazipur during April to June, 2012. Isolate no MY-1 (collected from Plant Pathology Division, BRRI, Gazipur) was used in this experiment. Effect of different Carbon sources on mycelial growth and sclerotia formation of R. oryzae-sativae - Carbon sources i.e. Dextrose, Sucrose, Galactose, Xylose, Fructose and Lactose were examined in this experiment. 2% Carbon source, 2% agar mixed with 100 ml potato extract and autoclaved. A 25 ml. of each treated source was poured into sterilized Petriplates (90 mm diameter). Treated Petriplates were inoculated aseptically with 6mm diameter culture block of mycelium obtained from 3 day old actively growing pure culture with the help of sterilized cork borer. Inoculated Petriplates were incubated at room temperature (27 ± 2ºC). Effect of different Nitrogen sources on mycelial growth and sclerotia formation of R. oryzae-sativae - Nitrogen sources i.e. Peptone, Yeast, KNO3, NaNO3, NH4NO3 and Urea were examined in this experiment. 2% Nitrogen source, 2% agar mixed with 100 ml potato extract and autoclaved. Mycelial disks of 6mm diameter were transferred from the margin of 3 day old colony in the centre of PDA plates and incubated at room temperature. Only PDA plates (without nitrogen source) served as control and compared with treated plates. Inoculated Petriplates were incubated at room temperature (27 ± 2ºC). Effect of different pH level on mycelial growth and sclerotial intensity of R. oryzae-sativae - Seven pH levels of 3.0, 4.0, 5.0, 6.0, 7.0, 8.0 and 9.0 were investigated for the growth of MY-1 isolates on PDA medium. The pH was adjusted with 50% lactic acid in the range of 3.0-6.0 and 10N NaOH in the range of 7.0-9.0 before sterilization. Mycelial disks of 6mm diameter were transferred aseptically from the margin of 3-day-old colonies at the centre of PDA plates (90 mm diameter) and incubated at room temperature. Data recording and data analysis - The mycelial colony diameter was recorded every day until the complete coverage with fungal growth in any plate. Four replicate plates were used for each treatment. Formation of sclerotia was also observed. Data were analyzed by CropStat (version 7.2) computer software.

The findings suggested that aggregate sheath spot might be favoured by potassium deficient soils. The pathogen R. oryzaesativae grew on PDA with maximum growth at 6.0- 9.0. Minimum growths were recorded at pH level 3.0- 5.0. The growth of this fungus reduced at very strong acidic and alkaline pH.
