Transfer of Wilt Resistant Gene(s) to Tomato (lycopersicon Esculentum Mill.)

Abstract/ Executive Summary:

The field trials of 1993-94 were carried out with twenty different lines/genotypes of tomato. Out of these tested materials, only one line V168 and two lines V167 and V168 were found to be resistant against bacterial wilt (Pseudomonas solanacearum) and Fusarium wilt (Fusarium oxysporum . sp. lycopersici) respectively. The rest lines were graded to be either moderately susceptible, susceptible or highly susceptible to the bacteria. In the case of Fusarium, 10 and 8 lines were recorded to be moderately susceptible and susceptible, respectively.

Objectives:

To develop wilt resistant variety in this country.

Methodology:

The plant materials comprised of 20 genotypes collected from the Tomato Breeding Project under Coordinated Genetic Research Project of the Department of Genetics and Plant Breeding, Bangladesh Agricultural University. Out of these genotypes, 10 were AVRDC lines and rest of them were recognized varieties of tomatoes. The experiments were conducted in a randomised block design with three replications at the experimental farm of the Department Genetics and Plant Breeding, BAU, Mymensingh. The rows were spaced 60cm apart with plant to plant distance of 50cm. The plot size was 2.5m x 1.8m for each genotype in each replication. The crop was grown on residual soil moisture and irrigation was provided whenever necessary. Proper drainage was also maintained. Recommended doses of fertilizers were applied to the soil and other intercultural operations were done to raise the crop.

Methods of inoculation: Root cutting method wilted plant affected by Pseudomonas solanacearum was collected from tomato field. Infected stem part was cut into small pieces and dipped in sterilized distilled water tor 4-6 hours. Bacterial ooze was liberated and this made the suspension milky white. This suspension was used as inoculum by root-cutting method where the roots of standing plants in the field were cut in three sides at 2-3 inches depth. the bacterial suspension was then poured onto the cut surface of roots uniformly and observations were made for onward development of the symptoms. During the experiment in the month of January, 1994 average temperature, relative humidity and rainfall were recorded to be 19.18°C, 73.32% and 51.00mm respectively. Leaf cutting method, upper young leaves of standing plants in the field were cut with a scissor which was dipped earlier in a suspension of 48 hours old 10 ml bacterial culture in a test tube mixed with 10 ml distilled water. In the control treatment, fresh sterile scissor was used to cut the leaves. Finally, all the treated plants were covered with polythene bags. Stem injury method six to seven week old healthy plants standing in the main field were inoculated by stem injury method. Individual plant was injured by a sterilized needle 5-6cm above the soil level and small mycelial block of 5-6 days old culture of Fusarium oxysporum f. sp. Iycopersici was placed on the injured area and wrapped with a wet sterilized cotton plug and sealed by a strip of cellophane tape. The whole plant was covered with a polythene bag. In the case of control plant, all the above practices were done except the use of mycelial block. This experiment was also carried out in the field during the month of January, 1994. Disease symptom was studied after 24, 48, 72 and 96 hours of inoculation and lesion development, if any, was measured with a centimeter scale.
