Study on Seed Priming of Watermelon and Bittergourd

Abstract/ Executive Summary:

The experiment was conducted at the laboratory and field of Seed Technology Division, BARI, Joydebpur, Gazipur, during January to March 2013 to determine suitable priming technique for improving seed germination and field emergence of watermelon and bittergourd. In case of watermelon, maximum germination percentage was obtained from hydroprimed seeds followed by osmoprimed and unprimed (control) seeds. Seedling vigour index was found maximum in hydroprimed seeds followed by unprimed seeds and the lowest vigour index was observed in osmoprimed seeds. Seedling emergence percentage in the field was also higher in hydroprimed seeds than that of osmoprimed and unprimed seeds of watermelon. In case of bittergourd, both hydropriming and osmopriming had no positive effect on seed germination, vigour index as well as field emergence percentage.

Objectives:

To determine suitable priming technique for improving seed germination and field emergence of watermelon and bittergourd.

Methodology:

The experiment was conducted at the laboratory and field of Seed Technology Division, BARI, Joydebpur, Gazipur during January to March, 2013. The experiment was carried out in a completely randomized design with four replications. In this experiment, seeds of watermelon var. Victor and bitter gourd var. Tia were used. Seed moisture content was determined by the high temperature oven method at 130⁰C for 4 hours. Mean moisture content of the seed sample of water melon was 7.33% and bitter goud was 7.26%. Seeds of watermelon were pretreated with thiram fungicide and bittergourd with provax in order to control possible fungal contamination during priming. Seed sample of each crop was divided into three sub-samples. One of the sub-samples was considered as control (unprimed) and other two sub-samples were prepared for priming treatments. In case of hydro priming, seeds of one sub-sample were soaked in distilled water and in case of osmopriming seeds of another sub-sample were soaked in solution of polyethylene glycol 6000 (PEG 6000) at a concentration of 253/litre distilled water giving an osmotic potential of -0.8 MPa (Michel and Kaufmann, 1973) for 12 hours. Priming treatments were incubated at ambient temperature under dark condition. After priming, seeds were removed and rinsed three times into tap water and then dried to at par original moisture level. Four replicates of 25 seeds from each sample germinated in sand media into cover plastic box to avoid moisture loss. Seeds were allowed to germinate at ambient room temperature for 14 days. At the end of germination test, germination percentage, root and shoot length, seedling dry weight were recorded. The vigour index was calculated according to Joyraj and Karivaratharaju, (1992). For field emergence, seeds were sown on raised bed at a depth of 3-5 cm with a spacing of 30cm5cm with four replications. After 14 days, healthy seedlings were counted. Data were analyzed statistically.

Achievement/Findings:

Result revealed that hydro priming is a simple and useful technique for improving vigour index as well as seedling emergence percentage of watermelon which may predict the stand establishment of the crop. Overall results of the study suggest that hydropriming is a useful technique for improving seed germination, vigour index as well as seedling emergence in the field of watermelon.
