Corn and Potato Starch as Cost Effective Alternative Gelling Agents for in Vitro Propagation of Orchid

Abstract/ Executive Summary:

An experiment was conducted at the appropriate growth conditions of shoot through in vitro propagation of orchid (Dendrobium sonia) using different gelling agents. Protocorms Like Bodies (PLBs) of orchid (one month oldprotocorms) were used for this study. Protocorms were cultured on MS medium supplemented with different concentrations of starches viz. corn (50 and 60gL^-1) and potato (50 and 60gL^-1) along with 2 gL^-1 agar used as alternative gelling gents as compared with 8 gL^-1 agar. The effect of shoot multiplication was observed at 35, 70 and 105 days after culture. The MS medium supplemented with the increasing dose of corn and potato gelled with 2 gL^-1 agar progressively increased their growth performance than that of 8 gL^-1 agar only. At the final stage, the best performance on shoots (number, length and fresh weight) and leaves (number, length and width) characters were recorded at 60 gL^-1potato. This study creates a new prospect of using cheap gelling agent which will reduce the production costs much and will help in popularizing plant tissue culture techniques.

Objectives:

This study creates a new prospect of using cheap gelling agent which will reduce the production costs much and will help in popularizing plant tissue culture techniques.
MS medium supplemented with different concentrations of starches viz. corn and potato at different stages at 35, 70 and 105 days to investigate their effects on shoot multiplication.

Methodology:

Protocorm Like Bodies (PLBs) of orchid (Dendrobium Sonia) were initiated and maintained in the Tissue Culture Laboratory of the Department of Biotechnology, Bangladesh Agricultural University, Mymensingh during the period from July to October 2009. These PLBs were used as plant materials. Treatments of starches like as corn (50 and 60 gL^-1) and potato (50 and 60 gL^-1) were supplemented into MS medium as alternative gelling agent of agar for in vitro propagation of orchid. One month old in vitro grown protocorms were placed in medium contained in 10 ml; vial. PLBs were cultured on 20 ml MS medium supplemented with different concentrations of starches viz. corn and potato at different stages at 35, 70 and 105 days to investigate their effects on shoot multiplication. The data for the characters under the present study were statistically analyzed following Completely Randomized Design (CRD).The analysis of variance was performed and means were compared by Duncan's Multiple Range Test (DMRT) at 5% level of probability for interpretation of results.

Achievement/Findings:

The potato starch is better than the other low cost gelling agent and more effective for plant tissue culture. So, the present study suggests the use of potato starch as an alternative gelling agent to agar reducing the production cost significantly.
