Evaluation of Multiple Ovulation and Embryo Transfer Technique in the Improved Production System in Black Bengal Goat in Bangladesh.

Abstract/ Executive Summary:

The aim of the research work was to confirm the results in terms of synchronization of oestrus using synthetic Prostaglandin F2α (GabbrostimR), adjust the dose of PMSG in terms of superovulatory response and refine the embryo collection procedure using surgical method of collection. Three MOET Flush programme was carried out during year 2. Like the Year 1 donors were synchronized using GabbrostimR (Alfaprostol, VETEM, Italy) at the dose rate of 2 mg equivalent to 1 ml/goat. Among 7 donors 6 donors responded (85.7%) following first time injection, where the mean time of onset of oestrus and duration of oestrus was 41.5± 8.11 (s.e.) h and 24 ± 4.51 (s.e.) h respectively. All 7 donors responded with second and third time injection, where the mean time of onset of oestrus and duration of oestrus was 43± 3.4 (s.e.) h, 41.1±1.92 (s.e.) and 27 ± 0.94(s.e.)h, and 28.1 ± 2.07(s.e.) h respectively. In all 3 MOET Flush super ovulation was performed using Folligon (PMSG, Intervet International B.V., Holland) at the does rate of 600 IU, 700 IU and 900 IU respectively. Only natural service was performed to fertilize the ovum. The superovulatory response was determined by counting the number of corpus lutea (CL) by mid ventral laparotomy. Only one donor responded in each case following 600 IU and 700 IU PMSG injections during the time of Flush I and Flush II. The total number of ovulation was 7 and 13 respectively. However all two donors responded following 900 IU PMSG injections during Flush III where total number of ovulation in each donor was 6 and 10 respectively. The embryos were collected by surgical means following the process as it was performed during the Year 1 work except the incision in this time was started more from the base of the teat. The embryos was collected and graded under stereomicroscope. The embryo collection rates in individual donor varied from 29 to 83 %. The percent of fertile embryos varied from 50-100%. In Flush I only one embryo was fertilized and it was Grade 2 morula. In Flush II among 8 recovered embryos 5 were Grade 2 morula and 3 were Grade 2 late morula. In Flush III from two donor goats among 10 embryos, 7 embryos were fertilized, 2 were Grade 1 late morula and 5 were Grade 2 morula. Effects of Gabbrostim during Year 2 time were consistent with the results of Year 1. 900 IU PMGS resulted better superovulatory response compared to 700 IU and 600 IU PMGS, which is in agreement with the results Year 1 work. However, the number of donor goats was very small during this year. The refinement of embryo recovery procedure during Year 2 flushing resulted more embryo recovery rate compared to year 1 work. No embryos were transferred because of unavailability of recipient goats.

Objectives:

1) To evaluate the MOET technique in Black Bengal Goat in Bangladesh

2) If the technique prove feasible in this breed future research will be conducted to speed up the process of genetic improvement of carcass quality including other economic traits for increasing the production system and economic development of the country.

3) This will also helps the students to understand the MOET technique practically.

Methodology:

Procedures was followed: 1) Synchronization of donors and recipients with prostaglandins 2) Superovulation of donors with superovulatory hormone 3) Natural service and /or AI of donors 4) Controlling of ovulation rate in donors through surgical procedure. 5) Flushing of the donors by surgical procedures to collect the embryos 6) Transfer of embryos to recipients by surgical procedure.

Achievement/Findings:

The refinement of embryo recovery procedure during Year 2 flushing resulted more embryo recovery rate compared to year 1 work. No embryos were transferred because of unavailability of recipient goats.
