Effects of Ga3 on Germination of Bitter Gourd Seeds

Abstract/ Executive Summary:

An experiment having eight concentrations of GA₃ {Control (tap water) , 10, 25, 50, 75, 100, 200 and 300 ppm} and three soaking time (6, 12 and 18 hr) was conducted at the laboratory of Plant Physiology Section of Horticulture Research Centre, Bangladesh Agricultural Research Institute during May 10, 2013 to July 16, 2013 to improve and enhance the germination of bitter gourd seed. The temperature and Relative humidity (RH) in the laboratory were 28±1°C and 75-80% during experimentation. The experiment was laid out in completely randomized design with three replications. Different soaking time, GA₃concentrations and their interaction showed significant variation in germination percentage, speed of germination, vigour index and plumule length. Eighteen hour soaking time and 10 ppm GA₃ independently or in combination gave the maximum germination percentage, speed of germination, vigour index and plumule length. Twelve hour soaking time coupled with 10 ppm GA₃ also produced results identical to those of 18 hour soaking time with 10 ppm GA₃ in respect of all parameters except speed of germination. GA₃10 ppm in combination with 12 or 18 hour soaking time proved better for germination of bitter gourd seeds.

Objectives:

To identify suitable concentration of GA₃ and soaking time for germination of bitter gourd seeds.

Methodology:

The experiment was carried out at the Laboratory of Plant Physiology Section, Horticulture Research Center, Bangladesh Agricultural Research Institute under ambient environmental conditions during May 10, 2013 to July 16, 2013. Within this period the experiment was repeated three times and average value of each data from repeated experiments was considered for analysis. The treatment consists of three soaking time (S₁ = 6, S₂ = 12 and S₃ = 18 hr) and eight GA₃ concentrations (T₀ = control, T₁ = 10, T₂ = 25, T₃ = 50, T₄ = 75, T₅ = 100, T₆ = 200 and T₇ = 300 ppm). The factorial experiment was laid out in completely randomized design (CRD) with three replications. The experiment was conducted using 72 earthen pots of equal size (22 cm top diameter and 6.5 cm depth) filled with equal quantity (2.30 kg) of coarse sand. Seeds were sown in sand at a depth of 2.5cm. Twenty five seeds were placed per pot. Slight watering was done immediately after sowing of seeds. Germination was started after 4 days of sowing. The temperature and Relative humidity (RH) in the laboratory were 28±1 °C and 75-80% during experimentation.Germination percentage and speed of germination were calculated.Vigour index: The 15 days old seedlings selected for calculating the seedling vigour index was oven dried at 72^oC for 72 hour. The oven dry weight of these seedlings was used as seedling dry matter. Seedling vigour index was determined by using the formula given by Abdul Baki and Anderson (1973) as below. Vigour index = Seedling dry matter x germination percentage(%). Ten seedlings of 15 days old were randomly uprooted from each pot. Then, after the separation of root from shoot, their plumule lengths were measured in centimeter. Finally, their mean had been used as the data related to the plumule lengths. The data collected were subjected to analysis of variance using MSTAT-C. Mean separation was performed by DMRT.

Achievement/Findings:

The findings of the study led to the conclusion that 10 ppm GA₃ with 12 - 18 hour soaking time was more effective in enhancing bitter gourd seed germination.
