Selection for Salt Tolerant Brassica Campestris Genotypes Through in Vitro Techniques

Abstract/ Executive Summary:

The experiment was conducted at Experimental field of Department of Genetics & Plant Breeding, Bangladesh Agricultural University, Mymensingh-2202. Ten Brassica campestris genotypes were selected and four salinity levels such as 0, 3, 6 and 12 dsm^-1(NaCl + KCl solution in a ratio of 1:1) were applied for screening at seedling stage against salinity. Seeds were germinated in (MS) medium containing different combination of salt solutions as mentioned above. To investigate the effect of different treatments, data on various parameters were collected and analyzed. All the considered parameters showed better performance in control and performances decreased with the increase of salinity level. However, at 12 dsm^-1most of the characters showed retarded performance. BARIsarisha-9, M-27-E2-1-5 and BARIsarisha-12 were tolerant to salinity as they showed better performance at higher salt level (6 dsm^-1) when parameters like germination percentage as 100%, 100% and 93.33 % respectively, total fresh weight was 12.6g, 36.2g and 10.8g respectively and total dry weight was 0.047g, 0.58g and 0.064 g respectively were considered. On the contrary, Agrani, Sampad and Tori-7 were more susceptible at the same salinity level. Estimation of Co-relation coefficient also indicated variable response of the individual genotypes for different characters in different salt concentration.

Objectives:

To investigate the effect of different levels of salinity on Brassica campestris.

Methodology:

The experiment was conducted at Experimental field of Department of Genetics & Plant Breeding, Bangladesh Agricultural University, Mymensingh-2202. This experiment was conducted for screening of 10 Brassica campestris genotypes for salt tolerance at seedling sage. 10 genotypes of Brassica campestris viz. Tori-7, Kallaynia, Sonali, Sampad, Agrani, BARIsarisa-9, BARIsarisa-12, BARIsarisa-14, BARIsarisa-15 and M-27-E 2-1-5 were used in the experiment. MS media containing 0, 3, 6 and 12 dsm^-1 (NaCl + KCl solution in a ratio of 1:1) were used to grow the seeds of the selected genotypes. Days to germination of seedling were recorded on the basis of regular observation and a seedling was considered as germinated when cotyledon of the seedling fully opened. Room temperature was 24° C with 16h day and 8h dark length and 100% humidity. The number of seedling emerged in each vial was recorded. Number of leaves per seedling were recorded when the age of the seedling were 25 days. The character was recorded on the 25^th day of sowing seeds. Analysis of variance was performed using the Plant Breeding Statistical Program. Simple correlation co-efficient between yield and different yield contributing characters were estimated.

Achievement/Findings:

The study revealed that BARI Sarisha-9, M-27-E 2-1-5 and BARISarisha-12 can withstand salinity to some extent. These genotypes can be brought under breeding program as potential parents for hybridization to produce salt tolerant genotypes.
