Temporal Variation in Seed Quality of Mung Bean Preserved in Different Seed Containers

Abstract/ Executive Summary:

An experiment was conducted to evaluate the seed quality of mungbean stored in different containers. Seeds of mungbean were collected from Agronomy department of Bangabandhu Sheikh Mujibur Rahman Agricultural University, Gazipur, Bangladesh in July, 2004. Immediately after collection, initial quality of mungbean seeds was assessed in the laboratory. Quality assessment in laboratory included germination, germination rate, germination index and coefficient of velocity. After initial quality assessment seed were kept in different containers like tin, polythene bag and gunny bag to observe the deterioration pattern. Among the three containers, seeds of gunny bag absorbed more moisture from surrounding atmosphere. The deterioration rate was also highest in seeds of gunny bag and the lowest seedling vigor.

Objectives:

To evaluate seed quality of Mungbean when stored in different containers.

Methodology:

Mungbean seeds were collected from Agronomy department of Bangabandu Sheikh Mujibur Rahman Agricultural University on July, 2004. Seeds were kept in three types of container such as tin pot, poly bag and gunny bag and stored at room temperature and RH for around 3 months. Collected seeds were compared for initial moisture content, seed size, germination and seedling vigor. Moisture content was determined by using oven dry method in the Agronomy laboratory of BSMRAU. 10 g of seeds from each 3 containers were taken and poured in a small container with cover and kept in an oven maintained at a temperature of 105⁰C for a period of 48 hours. The moisture content of seeds (wet basis) was determined by the following formula:

MC(%)= (M₂-M₃/M₂-M₁)x100, M₁= Wt. of container + cover, M₂ = Wt. of container + cover + mungbean seed before drying, M₃ = Wt. of container + cover + mungbean seed after drying.

Similar procedure was performed every fortnight from beginning of storage. Seed size of mungbean varieties were determined from the 1000 seeds counted from the pure seed by multi auto counter and weighed in an electronic balance. For germination test one hundred seeds of each sample were placed in Petri dish containing filter paper soaked with distilled water. For each test, four plastic pot were used. The plastic pots were placed in room temperature for 7 days for germination. Seedlings were counted every day up to the completion of germination at seventh day. A seed was considered to be geminated as seed coat ruptured, plumule and radicle came out up to 5 mm in length. Germination percentage was calculated using the following formula.

Germination (%)= (Number of seeds germinated/Number of seeds tested) x 100

Seeds were stored at room condition in different containers and germination test was performed every 10 days interval beginning from collection of seeds up to 50 days to observe the rate of natural ageing of mungbean seeds stored in different containers. Seedlings obtained from standard germination test were used for seedling evaluation. Normal or abnormal seedlings were classified according to the rules of ISTA. Seedling shoot and root length was measured on day 7 of the germination test. Ten plant samples from each plastic pot were harvested and shoot and root length of individual plant was recorded.
