Occurrence and prevalence of mango disease was studied in eight nurseries located at Dhaka, Gazipur, Barisal and Khagrachari district during July 2010 to April 2012 and three diseases were recorded during survey. The fungi anthracnose (Colletotrichum gloeosporioides) and leaf spot (Pestalotiopsis mangiferae) were identified firstly by symptoms observed and then confirmed by observing under compound microscope from infected leaves while sooty mould (Capnodium roseum) was identified only on basis of symptoms observed. Conidia of Colletotrichum gloeosporioides were dark coloured, single cell barrel shaped. Conidia of Pestalotiopsis mangiferae had three thickwalled, brownish, non-colors median cells and thin-walled hyaline apical and basal cells, fusiform and straight to slightly curve. Patches on leaf by forming a thin membranous covering over the affected parts was found on the sooty mold affected mango. Maximum incidence and severity of anthracnose and leaf spot was found at Khagrachari district and during the month of October respectively while maximum incidence of the sooty mold was found in January and Khagrachari district.

To study the seedling diseases of mango

 

The experiment was carried out in eight nurseries of Dhaka (Green orchid nursery, Agargaon and Barisal nursery, Savar), Gazipur (Gazipur nursery and Laxmipur nursery), Barisal (Sarchina nursery and Riyad nursery) and Khagrachari (Hill Research Center, Khagrachari and Ramghar nursery, Ramghar) during July 2010 to April 2012 to study the seedling diseases of mango. Three steps were followed in the experiment and these were i) Survey on seedling diseases of mango in selected nurseries; ii) Identification of causal organisms; iii) Epidemiology of disease incidence and severity. Survey on the seedling diseases of mango in some selected nurseries of Bangladesh: Age and number of seedlings: Age of mango seedlings and total number of seedlings in selected eight nurseries from July, 2010 to April, 2012. Observation of the symptoms: Symptoms of the diseases were studied by visual observation and disease was identified based on matching the observed symptoms in the infected plants with the symptoms published in Ber and other mango disease compendium. Identification of causal organisms: Diseased leaves were collected from the infected plants from represented surveyed areas and preserved in laboratory until isolation was made. Isolation of fungi were done by Moist blotter method and  Agar plate method. Purification: Fungi grown on medium were transferred to PDA plates following isolation of single hyphal tip technique. On PDA media Oidium magniferae and Cephaleuros virescense did not sporulate and for that matter it was not sub-cultured. Identification of the pathogen: Prepared slides were directly observed under compound microscope for identification of the associated organisms especially for Oidium mangiferae and Cephaleuros virescens. Attempts were also made to identify the fungal organisms grown out the inocula incubated in sterile wet filter paper by observing their growth characteristics under stereo-microscope and fungal organisms cultured on PDA media were also identified under compound microscope. Epidemiology of disease incidence and severity: Survey period: Altogether eight surveys were made in July 2010; October 2010; January 2011; April 2011; July 2011; October 2011; January 2012 and April 2012 respectively. Data collection during survey: 30 seedlings were randomly selected for counting diseased leaves and disease free leaves and moreover five leaves/plant were randomly selected to determine the disease severity. Plant infection, disease incidence and disease severity were determined. Collected data were analyzed statistically using MSTAT-C computer package program and mean was differentiated by Duncan’s Multiple Range Test (DMRT) at 5% level of significant.

 From the current study it can be concluded that the incidence and severity of anthracnose, leaf spot and sooty mold was varied by the variation of the location and time due to the variation of the environmental conditions like temperature, rainfall, relative humidity in different locations and time of Bangladesh.
