Dr. Syed Sakhawat Hussain
Professor
Department of Animal Breeding and Genetics, BAU, Mymensingh-2202.

The experiment was conducted at the Artificial Insemination Laboratory under the Department of Animal Breeding and Genetics, Bangladesh Agricultural University, Mymensingh and the surrounding area of the Bangladesh Agricultural University, campus. The present study was conducted to observe the efficiency of preserved semen for AI to rural goat population and compare the growth performance of progeny by using Artificial Insemination between selected and random buck. The parameters of semen characteristics were included semen volume (ml/ejaculate), sperm concentration (million/ml), sperm motility (%), live spermatozoa (%), normal spermatozoa (%) pH of raw semen and sperm motility (%), live spermatozoa (%), normal spermatozoa (%) of diluted semen at 1st, 2nd and 3rd day of preservation. Data of growth performance of progeny were collected from the project area. Data were analyzed using SAS and VCE computer program. The selected buck had significant effect on average semen volume (ml/ejaculate), sperm concentration (million/ml), sperm motility (%), live spermatozoa (%), normal spermatozoa (%), but had no significant effect on semen pH . The highest semen volume sperm concentration, sperm motility (%), live spermatozoa (%), normal spermatozoa (%) and pH were found to be 0.51 ± 0.23, 2345 ± 84.10, 82.80 ± 3.43, 85.62 ± 0.57, 87.53 ±1.42 and 6.6 in selected buck number 1439; and the lowest semen volume, sperm concentration (million/ml), sperm motility (%), live spermatozoa (%), normal spermatozoa (%) and pH were found to be 16 ± 0.05, 2023 ± 67.60, 70.00 ± 5.54, 82.62 ± 2.13, 85.32 ± 0.72 and 6.6 in selected buck number 118. For study of diluted preserved semen the data clearly indicated that when dilute semen was storage up to 3 days, the sperm motility, percentage of live spermatozoa and percentage of normal spermatozoa. The highest significant (p<0.05) effect was found incase of percentage of motility, percentage of live spermatozoa and percentage of normal spermatozoa with different period (1st, 2nd and 3rd day) in selected and random bucks. The selected bucks had significant effect on birth weight, 3 month weight and 6 month weight. The selected bucks had significant effect on conception rate. The highest (1.40 ± 0.11 kg) and lowest (0.99 ± 0.10 kg) birth weight were shown by selected buck number 1439 and random buck number 118 respectively. The heritabilities for all the traits studied were high, ranging from 0.62 ± 0.06 to 0.88 ± 0.06 indicating that genetic improvement in these traits through selective breeding seems to be promising. The selected bucks had no significant effect on conception rate. Average conception was found to be 50.32%. It was the first attempt in the country to use dilute semen through AI. Selected bucks were found to be superior over random bucks in terms of semen quality and performance of progenies.

Buck, semen, rural goats, optimum fertility, phenotypically

  Animal Health and Management
  Goat

1) Collection of superior bucks selected phenotypically with their body weight and daily gain.

2) Evaluation of diluted semen for maintaining their keeping quality and optimum fertility

3) Study the conception rate with AI in does around the BAU campus and familiarize AI in goats to the rural farmers for the genetic improvement of goat population.

4) Selection of superior bucks produced out of the AI services and distributed them to the rural farmers for their economic gain.

1) Material and procedure: In the beginning of the project 6 mature Black Bengal bucks will be procured at the age of 6 month following the previous selection criteria. The bucks will be maintained at the Artificial Insemination Laboratory under the Department of Animal Breeding and Genetics, Bangladesh Agricultural University, Mymensingh. All the bucks will be housed and managed appropriately in very good hygienic condition. The bucks will be ear tagged. All the bucks will be trained to collect semen using Artificial Vagina (AV). Semen will be collected thrice in a week. Just after collection semen will be evaluated for its quality such as colour, volume, pH, consistency, motility, concentration, morphology and percent of live spermatozoa. After completion of macroscopic and microscopic evaluation of semen the semen sample will be diluted with EYC diluter. The combination of EYC would be as follows: Egg yolk = 25 ml Sodium citrate buffer (2.94%) = 50 ml Penicillin = 1000iu/ml Dihydro streptomycin = 1000 mg/ml Sulphanelamide = 0.6gm/100ml (a) Parameter for raw semen: - Volume - Colour - Motility - Concentration - Morphological tests (b) Parameter for diluted semen: - Motility (%) - Live spermatozoa (%) - Normal Spermatozoa (%) (c) Parameter for AI work: - Conception rate of 1st, 2nd and 3rd day service with preserved semen - Number of service per conception - Gestation length - Body weight of kids produced - Monthly body weight - Body weights gains as birth to 3-month, 3 to 6-month, 6 to 9-month and 9 to 12 month - Age at first heat - Weight at puberty - Age at first conception.

(b) Design of experiments: The recorded data of the project will be stored in computer and enter twice in a week under the different elements of goat data file. The nature of data will presumably be on various animals of different ages, sex, birth types, season and groups. The total number of experimental animals will also vary among different groups and locations. As such the design of experiment will essentially be non-orthogonal or unbiased factorial in nature. The collected data will therefore, be analyzed using General Linear Model (GLM) procedure of Harvey Mixed Model Least Squares and Maximum Likelihood computer package (Harvey, 1990) and SAS and data will be plotted and arranged by GenState (2002). The least squares analysis of variance (LSANOVA) will be performed for all the traits and Least Squares Means (LSM) will be calculated and compared by LSD tests.
