Sword suckers of BARl Kola-3 and BARI Kola-4 were collected from ARS, Raikhali, Chandragona, Rangamati Hill District. Shoot tips were separated from those suckers and cultured on Murashige and Skoog (MS) medium supplemented with different concentrations (viz. 1, 2, 3, 4, 5 and 6 mg/1 BAP) of BAP and zip (viz. 1.0, 1.5 and 2.0 mg/l) to optimize existing protocol for in vitro production of BARl Kola-3 and BARl Kola-4. Among the different concentrations, MS medium supplemented with 3 mg/1 BAP was found suitable for in vitro production of multiple shoot from in case of BARl Kola- 4, 5 mg/l BAP was found suitable for in vitro production of multiple shoots. Both of the varieties produced well developed roots in 1/2 MS medium supplemented with 0.5 mg/1 IBA. Between the varieties, BARI Kola-4 showed the better performance compare to BARl Kola-3 for in vitro propagation. After 05 days hardening at room temperature followed by 07 days under polythene house hardening, 100% transplanted plantlets survivals which were allowed 1.5-2.0 months for planting at field. Fifty validation trials had been established at farmer's field along with two on-stations at eight upazillas of Khagrachari and Rangamati Hill Districts. Tissue cultured BARI kola-3 produced 50% flowering at 360 days on 22.13 leaves stage followed by BARI kola-4 (343 days) at 22.67 leaves stage. Approximately 32 days earlier harvesting had been recorded at BARI kola-4 than BARl kola-3. BARl kola-4 produced 49.95 t/ha yield followed by 47.25 t/ha in BARI kola-3. Tissue culture plantlets of BARl kola-3 and BARl kola-4 produced 13.20% and 18. 92% higher yield, respectively compare to traditional sucker. Moreover, March-April plantations gave 7.82% and 9.18% higher yield, respectively in BARl kola-3 and BARI kola-4 than September-October plantation. Sword suckers of BARI Kola-3 and BARI Kola-4 were collected from ARS, Raikhali, Chandragona, Rangamati Hill District. Shoot tips were separated from those suckers and cultured on MS (Murashige and Skoog, 1962) medium supplemented with different concentrations (viz. 1, 2, 3, 4, 5 and 6 mg/I BAP) of BAP and zip (viz. 1.0, 1.5 and 2.0 mg/I) to optimize existing protocol for in vitro production of BARI Kola-3 and BARI Kola-4. The experiment was designed as CRD having 5 replications where each replication contained 10 jars. The inoculated cultures were incubated at 25 ± 1°C under approximately 2500-3000 lux by cool white fluorescent tubes for 16/8 h light/dark cycle in 60-70% relative humidity. Data on percentage of success, bud initiation, no. of shoots /culture, length of shoots, days to root initiation, no. of root and length of roots/plant etc. was recorded. Before planting, randomly 100 sample plantlets were tested for bunchy top virus using Agdia Bunchy top Virus Testing Kit (USA). After successful standardization of protocol for in vitro production of BARI Kola-3 & 4, 18000 plantlets were produced for validation trials at different locations of Khagrachari and Rangamati hill districts. Fifty validation trials was established at 08 upazillas of Khagrachari and Rangamati hill districts. Data on different parameters like base girth, plant height, no. of leaves, days to 50% flowering, days to harvest, yield, insect and disease infestation etc. was recorded.

MS medium supplemented with 4 mg/I BAP was found suitable for in vitro production of multiple shoot from the shoot tip of sword sucker of BARI kola-3. In case of BARI kola-4, 5 mg/l BAP was found suitable for in vitro production of multiple shoots. 1/2 MS medium supplemented with 0.5 mg/l lBA has been found suitable for in vitro root production for both of the varieties. After 05 days hardening at room temperature followed by 07 days under polythene house hardening, plantlets should be transplanted at polybags and allowed 1.5-2.0 months for transplant at field. Hill valleys are suitable for tissue cultured banana production followed by hill base where have minimum irrigation facilities. March-April plantation has been found suitable for banana cultivation at hilly areas. No bunchy top virus infected plantlets were found at nursery as tested by Agdia Bunchy top Virus Testing Kit (USA). Farmers can produced quick growing vegetables like lalshak, radish, pumpkin, tomato, cotton, bush bean etc. as inter crop in the tissue cultured banana field during the vegetative growth stage.
