Validation Trial of Tissue Cultured Plantlets of BARI Maslta-1

Abstract/ Executive Summary:

Tissue cultured and grafted saplings of BARl malta-1 was planted to see the field performance at ARS. Ramgarh and ARS, Raikhali. Maximum plant height (225 cm) and number of brunches (8.5) were recorded in tissue cultured plants where base girth was 22.22 cm. But highest plant canopy N-S= 292.5 and E-W= 292 cm was found in grafted plants where tissue culture plants have N-S= 175 cm and E-W= 180.75 cm. Vigorous growth and development had been seen in tissue cultured plants compare to grafted ones up to second year and plant canopy was found higher in tissue cultured plants (N-S = 111. 75 cm, E- W = 89.25 cm) compare to grafted ones (N-S = 91.25 cm, E-W = 89.5 cm). Highest plant height (199.25 cm), base firth (10.5 cm) and no. of branch (8. 5) also found in second year tissue culture plant followed by grafted plants. After second year grafted plants shown vigorous growth and development. Both of tissue cultured and grafted plants produced flowers after three years but only grafted plants have carried few fruits

Objectives:

Objective of this study was to see the field performance of tissue cultured plantlets of BARl Malta-1 compared to grafted one.

Methodology:

The regenerated rooted plantlets with 5-6 leaf stage were transferred from culture room and kept in room temperature (30°C) for 5-7 days. The plantlets were then removed from the culture tubes and all the adhering media were carefully washed out so that, the root damage was the least. Washed plantlets were planted into small polyethylene begs containing sterile soil, sand and decomposed cow dung at the ratio of 1: 1: 1. Subsequently, the healthy plants were transplanted in a 9 x 12 inch earthen pots containing soil mixture at nursery for three months. Well established saplings were transplanted along with grafted one at Hill Agricultural Research Station, Raikhali, Rangamati Hill Districts and Hill Agricultural Research Station, Ramgarh, Khagrachari Hill Districts in May, 2011. Saplings were transplanted at single line design following plant to plant distance 4.0 m. Single plant was considered as one replication. Data on Plant height, base girth and number of branches/plant were recorded.

Maximum plant height (225 cm), no. of brunches (10.0) was found in tissue cultured plants where base girth was 22.22 cm. Up to second year plant canopy was found higher in tissue cultured plants ( N-S = 118.25 cm, E-W = 109.25 cm) compare to grafted ones (N-S = 88-31 cm, E-W = 79.67 cm). However, in second year vigorous growth and development was found in Tissue cultured plants compare to grafted ones. In case of ARS, Ramgarh maximum plant height (199.25 cm), no. of brunches (8.50) was found in tissue cultured plants where base girth was 10.5 cm in tissue cultured plants. Plant canopy was also found higher in tissue cultured plants (N-S = 111.75 cm, E-W = 89.25 cm) compare to grafted ones (N-S = 91.25 cm, E-W = 89.5 cm. However, vigorous growth and development has been found in Tissue cultured plants compare to grafted ones during second year. There was no flowering in tissue cultured and grafted plants up to second year. A significant result has not yet been generated. So, the investigation should be continued for the next years.
