Detection of Rb Gene in Selected Transgenic Late Blight Resistant Hybrid Clones Through Pcr

Abstract/ Executive Summary:

Six selected transgenic late blight resistant hybrid clones were used to find out the RB gene integration. DNA was extracted from 6 clones along with non- transgenic control variety Diamant and Cardinal. Through PCR method RB gene confirmation test was done. Among the 6 hybrid clones, 5 showed positive bands which indicated that RB gene were present of those clones.

Objectives:

Objective of the study was to select late blight resistant hybrid clones that can be released as commercial varieties in future.

Methodology:

Six hybrids clones along with control variety Diamant and Cardinal were used in this study isolation of genomic DNA was done from fresh leaf samples. N terminal primers were used. DNA amplification by PCR was done using Thermal Cycler. The amplified DNA products were separated by electrophoresis in 1% agarose gel.

Using N terminal primer, among the six hybrid clones, five showed positive bands with RB gene and one showed negative.
