Studies on the Genetic Diversity of Okra (abelmoschus Esculentus L. Moench.) Using Peroxidase Isozyme

Abstract/ Executive Summary:

The experiment was conducted with 116 okra genotypes using polyacrylamide gel electrophoresis in the Department of Horticulture, Bangladesh Agricultural University, Mymensingh. There were eight different electrophoretic zymotypes was observed, all the zymotypes were polymorphic means presence of more than one band in each genotype. The genotypes were grouped in different polymorphic zymotypes, indicating considerable genetic diversity among the studied okra genotypes. Zymotypes frequency of peroxidase isozyme was maximum in cluster III followed by cluster IV, cluster I, cluster VI, cluster II, cluster VI and cluster VIII. The lowest zymotype frequency was in cluster IV. There are 8 electrophoretic zymotypes were observed and formed 29 bands at different Rf values varied from 0.05-0.42. Peroxidase band in 0.05 Rf values is constant but other bands were variable in different Rf’s. Similarity coefficient between different banding pairs was observed significantly different in studied okra genotypes. Zymotypes of higher frequency are the representative of less variation and the lower frequency of the genotypes in different zymotypes indicated random distribution of okra genotypes throughout the country.

Objectives:

To estimate the genetic diversity of okra using peroxidase isozyme and selection of genotypes for hybridization program

Methodology:

The experiment was undertaken with 116 okra genotypes using polyacrylamide gel electrophoresis (PAGE) techniques (Staub et al. 1997). Peroxidase isozyme has been tested for activity of enzymatic proteins extracted from young leaves of 25-30 days old seedlings of okra. Peroxidase isozyme analysis was done in the biotechnology laboratory, Department of Horticulture, Bangladesh Agricultural University, Mymensingh during 2005-2006. Banding patterns of peroxidase isozyme was recorded on the basis of number and the relative front (Rf) values of the bands following Mouemar and Gasquez (1983). The Rf value of each respective band on schematic isozyme pattern was determined to allow precise comparisons among the various genotypes. For estimating the relationship between electrophoretic phenotype similarities coefficients were calculated using index of genetic similarity comparisons (S_xy), which were:
S_xy = 2n_xy/ (n_x+n_y)
Where, 2n_xy = number of shared bands,
n_x = number of bands in electrophoretic patterns of x, and
n_y = number of bands and electrophoretic patterns of y.

Similarity coefficient (Sxy) values of electrophoretic phenotypes were subjected to cluster analysis employing the unweighted pair group method using arithmetic means (UPGMA). Preparation of dendrogram and cluster analysis, the value 1 was put for the presence of the electrophoretic pattern and value 0 was used against the absence of the pattern for each genotypes. The original data was transformed to Z-scores prior to cluster analysis. Zymotypes were used for clustering and the Euclidean distance method was used for the dissimilarity.

Achievement/Findings:

It is concluded that there exists a wide range of variability among the okra genotypes. According to the clustering on the basis of genetic distance of the genotypes, the parental plants might be selected from different clusters; it was much more likely to get the desirable characters for F₁ generation for hybrid development.
