Study of Growth Performance and Genetic Variation of Thai and Local Koi.

Abstract/ Executive Summary:

Koi (Anabas testudineus) is widely distributed in Indo-Pak-Bangladesh sub-continent to China but the culture potentials are heterogenic. For this why, an experiment was undertaken to study the growth performance and genetic variation of Thai and local koi. In this regard, for growth analysis, 24 days of old larvae were collected from “Brahmaputra Fish Seed multiplication farm” and stocked in earthen ponds (3 ponds for each source) at stocking density of 250 fish/pond and reared for 3 months using supplementary feed. However, Thai koi (82.93±0.76 g) showed almost double growth than the local koi (43.90±0.50 g). On the other hand, for genetic analysis, cross koi with Thai and local koi were used. In this instances, ten allozymic enzymes (CA 6.1 & TC-1 buffer system) were screened those were encoded by eleven enzymatic loci (Adh-1, Est-1, G3pdh-1, Idhp-1, Ldh-1, Mdh-1, Mdh-2, Pg.n, Sdh-1, Gpi-1 and Gpi-2). Four loci Est-1, G3pdh-1, Gpi-1 and Mdh-1 were showed polymorphic (p˂0.95) in all the populations. The genetic variabilities (mean proportion of polymorphic loci, mean proportion of heterozygous loci per individual, mean of number alleles per locus, the observed and expected heterozygosity are 36.36%, 13.333%, 1.636, 0.227 and 0.144 respectively) were higher in cross koi rather than Thai & local koi. In UPGMA dendogram, local koi population made one cluster and differentiated from other populations by the genetic distance D= 0.142. Therefore, in growth analysis Thai koi showed highest growth and in genetic analysis, cross and Thai populations are formed one cluster that indicates that cross koi might have important implication for our aquaculture production.

Expected Output: 1.The sustainable culture of the local and Thai koi will be carried out by the growth study of them. 2. Low cost diet will be produced to minimize the culture cost. 3 The morphological variation will be identified. 4. The genetic structure of both koi will be analyzed which will be helpful for identifying them.
