In order to investigate the protective role of polyamines (PAs) in maize under drought stress, maize seedlings (Zea mays cv  Khoibhutta) were imposed to 20% polyethyleneglycol (PEG) containing 50 μM of putrescine (Put), spermidine (Spd) and spermine (Spm) for 48 hours and the contents of relative water content (RWC), proline, chlorophyll and malondialdehyde (MDA), hydrogen peroxide (H2O2), rate of superoxide radicals (O2•-) generation and methyl glyoxal (MG) and the antioxidant enzyme activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), ascorbate peroxidase (APX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), glutathione S-transferase (GST), glyoxalase I (Gly-I), glyoxalase II (Gly-II) and lipoxigenase (LOX) were checked. Treatment of Put, Spd and Spm effectively maintained the balance of water content and chlorophyll contents in leaves. In addition, the data indicated that the pretreatments reduced accumulation of the proline suggesting the reduction of stress effect. They also decreased H2O2 content, rate of O2•- generation, and prevented drought induced MDA. The activities of CAT, APX and GPX were found to increase in response to Put. Treatment of Put, Spd and Spm increased MDHAR activity, contrary they reduced LOX activities remarkably. Therefore, the antioxidant could be changed by PAs, which had the ability to modulate the radical scavenging system and to lessen  the oxidative stress in this way. The results suggested that, pretreatment with Put, Spd and Spm prevents oxidative damage, and the protective effect of Put was found to be greater than that of Spd and Spm.

To examine the effects of exogenous polyamines (PAs) under polyethyleneglycol (PEG) induced drought stress in maize

Eight day old seedlings of maize (variety Khoibhutta) grown in petridish were imposed to drought by adding 20% PEG in incubator. Fifty μM of Put, Spd and Spm were maintained in the PEG solution. Control plant with and without PAs were maintained with same condition. Data were taken after 48 h of stress implementation. Leaf relative water content (RWC) was measured according to Barrs and Weatherley (1962). Fresh weight (FW), turgid weight (TW) and dry weight (DW) of leaves were recorded. The RWC was calculated by the following formula: RWC (%) = (FW-DW)×100/(TW-DW). Data were analyzed by SAS (Version 9.1) program following complete randomized design (CRD) and the mean differences were compared by least significant difference (lsd) test. Values of mean ± SE were calculated from three independent experiments and P<0.05 was considered to be significant.

Considering all, it could be concluded that, exogenous application of PAs maintained the chlorophyll and RWC in maize leaves and reduced the contents of ROS. Treatments of PAs reduced accumulation of the proline suggesting the reduction of stress effect. The antioxidant could be changed by PAs, which had the ability to enhance the radical scavenging system by modulating enzymatic and non-enzymatic antioxidants and to lessen in this way the oxidative stress. The results suggested that, the treatment with Put, Spd and Spm prevented oxidative damage, and the protective effect of Put was found to be greater than that of Spd and Spm.
