Evaluation of Nitrate Reductase Activity, Leg-hemoglobin and Chlorophyll Contents in Chickpea

Abstract/ Executive Summary:

A pot experiment was conducted at BINA Mymensingh during November 1993 to February 1994 with 50 exotic germplasm of chickpea to determine their nitrate reductase (NR) activity and chlorophyll contents. With another field experiment, 24 mutants/progenies were evaluated for leg-hemoglobin contents in addition to chlorophyll and NR activity. CRD and RCBD were used for pot and field experiment respectively. NR activity ranged between 0.30 to 1.84 µmol NO₂^-/gfw/h. However, 35.3% of the exotic genotypes had above 1.0 NO₂^-/gfw/h NR whilst 64.7% showed below 1.0. All the mutant lines showed higher NR activity than that of their mother G-97 excepting L-67 and L-62. All the F₆ progenies showed lower NR activity than that of their lowest NR activated parent. Total chlorophyll contents ranged from 0.68 mg/gfw to 2.18 mg/gfw among exotic germplasm. Additionally, chlorophyll a/b ratio in them also varied from 0.89 to 1.32. In contrast, total chlorophyll content in mutants/progenies ranged from 2.32 to 2.78 mg/gfw. Of the progenies and mutants P54, P-66, L-88 showed higher chlorophyll contents as compared to the rest. Leg-hemoglobin content among the mutants/progenies ranged from 6.36 to 11.76 mg/gfw and G-97, L-37, Hyprosola, G-319 and P-66 being higher than all those rest.

Objectives:

Methodology:

A pot experiment was conducted at BINA Mymensingh during November 1993 to February 1994 with 50 exotic germplasm of chickpea, collected from ICRISAT to determine their nitrate reductase (NR) activity and chlorophyll contents. With another field experiment, 24 mutants/progenies were evaluated for leg-hemoglobin contents in addition to chlorophyll and NR activity. Seeds were sown in earthen pots for the 1st experiment containing 8 kg soil. The soil of each pot was fertilized with recommended doses of PK. Ten seeds of each genotype were sown and fifteen days after germination they were thinned out of 5 plants. N-free complete nutrient solution was applied twice in a week. Other cultural practices were followed when necessary. In the 2nd experiment 24 M₆ mutants/F₆ progenies were directly grown in the field using a randomized complete block design with three replications. Recommended doses of fertilizer were applied at land preparation. Fifty day old plant leaves were assayed for NR after 48 hours of application of KNO₃ @ 10 mM. Chlorophyll was also determined for the genotypes at the age of 52 days. Leg-hemoglobin, NR and chlorophyll contents of the 2nd experiment were assessed at the age of 65 days.
