In Vivo Nitrate Reductase Activity in Inoculated and Non-inoculated Chickpea in Relation to Reductant Energy Supply

Abstract/ Executive Summary:

An experiment was conducted two chickpea genotypes viz. G-299 and G-97 at BINA, Mymensingh during February to June 1993. Seeds of chickpea genotypes were sown in Leonard jars containing sands. Seeds were surface sterilized by 50% ethyl alcohol and 3% H₂O₂ for 3-4 minutes followed by 7 times washing with sterilized water and sown in sands and in it mixed inocula (BINA-CP-11 + BINA-CP-12 + BINA-CP-19) were provided at the crown root zone of the seedlings. All the plants were supplied with 5 mM nitrate in addition to nutrient solution at the age of 30 days and analysis for NR were done at 24, 48 and 72 hours after NO₃^-addition. Varietal differences in the induction of NR was observed, NR was maximally included in 24 hours in G-299 whereas G-97 showed a delayed induction at 48 hours. At 72 hours, G-299 maintained a higher activity than G-97. In G-299, activity of NR was more in nodulated chickpea than that of non-nodulated chickpea except at 24 hours. In G-97, nodulated and non-nodulated chickpea showed similar activity of NR up to 48 hours but at 72 hours, high activities were reported in nodulated chickpea.

Objectives:

Objective of the study was to explore the energy status in chickpea with respect to simultaneous operation of both nitogenase and nitrate reductase systems of N-assimilation.

Methodology:

An experiment was conducted two chickpea genotypes viz. G-299 and G-97 at BINA, Mymensingh during February to June 1993. Seeds of chickpea genotypes were sown in Leonard jars containing sands. According to Hewitt (1966) all the Leonard jars including nutrient solution and sand were sterilized by autoclaving for 20 minutes at 15 psi (121^oC). Sterilized nutrient solutions were added every three days. Seeds were surface sterilized by 50% ethyl alcohol and 3% H₂O₂ for 3-4 minutes followed by 7 times washing with sterilized water and sown in sands and in it mixed inocula (BINA-CP-11 + BINA-CP-12 + BINA-CP-19) were provided at the crown root zone of the seedlings. All the plants were supplied with 5 mM nitrate in addition to nutrient solution at the age of 30 days and analysis for NR were done at 24, 48 and 72 hours after NO₃^-addition. The NR analysis was done following method of Stewart and Orebamjo (1979). In the assay solution glucose, sucrose and lactose were provided to test the effect of those carbohydrates for NR activity. For a blank, assay solution without carbohydrates and leaf material was used.

Achievement/Findings:

The enhanced activity of nitrate reductase in presence of added carbon sources both under nodulated and non-nodulated conditions showed that chickpea, either nodulated or non-nodulated had initiation of reductant energy for nitrate assimilation.
