Screening of Chickpea Genotypes for High Efficiency of N-utilization

Abstract/ Executive Summary:

The experiment was conducted with 45 chickpea genotypes at BINA, Mymensingh during November 1991 to February 1992 to identify chickpea genotypes for their status of nitrate reduction capacity among the existing genotypes. In chickpea, nitrate reduction capacity as expressed by nitrate reductase activity was high and the values were among 45 genotypes between 1.39 to 7.32 units with an average of 3.79 units and a variation of ± 1.48 units. Thirty five percent of the total genotypes showed NR activity more than 4 units. The results suggested that productivity of chickpea could be increased by combined N sources by proper selection of genotypes on the basis of NR activity.

Objectives:

Objective of the study was to identify chickpea genotypes for their status of nitrate reduction capacity among the existing genotypes

Methodology:

The experiment was conducted with 45 chickpea genotypes at BINA, Mymensingh during November 1991 to February 1992. All the chickpea genotypes were collected from Plant Breeding Division, BINA, Mymensingh. The plants were grown in sand taken in the pots culture under the glass house condition. The plants were regularly supplied with 0.25 strength Long Ashton (Hewitt, 1966) nutrient solution without N in the medium up to the date at which nitrate was provided @ 5.0 mM at 48 or 72 hours before the analysis of Nitrate reductase (NR). For the analysis of NR, 100 mg of selected leaf samples taken in a scintillation vial with 5.0 ml of freshly prepared assay solution containing 0.1M phosphate buffer of pH 7.5, 1.5% N-proponol, 0.05% Neutronix and 1.5% Potassium nitrate were vacuum infiltrated in a desiccator and dark incubated for 1 hour at 27^oC with moderate shaking. After 1 hour, 1.0 ml of extract was added to 2.0 ml of color reagent which contained 1% sulphanilic acid and 0.02% NEDD. Blank was prepared with 1.0 ml of assay solution with 2.00 ml of color reagents. The NR activity was expressed as µmol NO₂ /g fresh weight /hour.
