The experiment was conducted with 29 peanut genotypes at BINA, Mymensingh during January to May 1992 to identify peanut genotypes for their status of nitrate reduction capacity among the existing genotypes. In peanut, nitrate reduction capacity as expressed by nitrate reductase activity was high and the values were among 29 genotypes between 1.20 to 3.24 units with an average of 2.24 units and a variation of ± 0.56 units. NR activity in all the peanut genotypes was found low (<4.0).

To identify peanut genotypes for their status of nitrate reduction capacity among the existing genotypes

The experiment was conducted with 29 peanut genotypes at BINA, Mymensingh during January to May 1992. All the peanut genotypes were collected from Plant Breeding Division, BINA, Mymensingh. The plants were grown in sand in the pots under the glass house condition. The plants were regularly supplied with 0.25 strength Long Ashton (Hewitt, 1966) nutrient solution without N in the medium up to the date at which nitrate was provided @ 5.0 mM at 48 or 72 hours before the analysis of Nitrate reductase (NR). For the analysis of NR, 100 mg of selected leaf samples taken in a scintillation vial with 5.0 ml of freshly prepared assay solution containing 0.1M phosphate buffer of pH 7.5, 1.5% N-propanol, 0.05% Neutronix and 1.5% Potassium nitrate were vacuum infiltrated in a desiccator and dark incubated for 1 hour at 27oC with moderate shaking. After 1 hour, 1.0 ml of extract was added to 2.0 ml of color reagent which contained 1% sulphanilic acid and 0.02% NEDD. Blank was prepared with 1.0 ml of assay solution with 2.00 ml of color reagents. The NR activity was expressed as µmol NO2 /g fresh weight /hour.

NR activity in all the peanut genotypes was found low (<4.0), so induction of variability for higher activity of NR was suggested.
