The research was performed to estimate the prevalence of subclinical mastitis in cows kept in Bonoful dairy farm and other mini dairy farms at Anwara of Chittagong district in Bangladesh using California Mastitis Test kit (C.M.T tester). Investigation was carried out on 153 milk samples, 100 samples of them revealed positive reaction that denote the cows were suffering from subclinical mastitis (65.5%). In this study, 113 milk samples from crossbred dairy cows and 40 samples from Red Chittagong cows. From the 113 samples 95 were (+ve) that is (84%) and rest 40 samples from Red Chittagong cows and only 5 were (+ve) that is 12.5%. Bacteriological examination for isolation of causative organism was done using those samples which showed strong positive result (+++) in CMT test. In this study, the cows were mostly infected with mixed infection Staphylococcus spp. and E. coli and some cows had mono bacterial infection.

To detect the prevalence of subclinical mastitis and isolation of causal agent in lactating cows in Bonoful Dairy farm and mini dairy farms at Anwara, Chittagong.

A total of 153 dairy cows were selected for this study and were sampled during the period of March 2013 to February 2014 from BDF (Bonoful Dairy Farm) and different private sector mini dairy farms at Anwara, Chittagong (Number of cows per farm 3 to 40). In this study, only apparently healthy crossbred and local Red Chittagong cows were used. In this study, 40 samples were collected from BDF, Anwara Chittagong. Those cows were crossbred cows and other 113 cows were Red Chittagong and cross breed of LF and LFS. Before collection of milk the teat and tips were washed with clean water, antisepsis was done with a swab soaked with 70% alcohol and then milk sample were collected aseptically from the udder at the time morning milking. All the milk collected vials were labeled with identification numbers of cows or identifying symbol of cows.

 Immediately after collection of milk sample were subjected to physical examination with naked eyes to detect any abnormalities in color, consistency and presence of any other clot blood flakes and any other visible abnormalities. For detection of subclinical mastitis the procedure was followed in this study as per manufacturer’s instruction.

 Before incubation the sample was allowed in normal temperature. Then 100μL of milk sample was taken with micro pipette and the sample was expelled on a test-tube containing 10 ml Peptone broth. These works were performed within a Laminar air flow for aseptic measure. Then the milk sample containing peptone broth was incubated at 37°C for 24 hours. After incubation one loop of incubated sample was striked on EMB agar, Mac Conkey agar, Brilliant green agar and mannitol salt agar respectively and again incubated at 37ºC for 24 to 48 hours. The bacterial isolates were identified by their cultural, morphological and biochemical characters. For the cultural characteristics, discrete colonies on the agar surface were observed. Their shape, size, consistency and color were studied. Gram’s stained slides of the isolates were examined microscopically to study their cellular morphology. The biochemical tests were performed as fermentation with five basic sugars (dextrose, maltose, lactose, sucrose and mannitol) and thereby production of acid or gas or both. Individually isolated colonies of the same morphology were selected from appropriate agar plates, cloned and checked for purity of growth prior to characterization of the respective genera and species. Characterization of the respective genera were done on the basis of morphological, cultural and biochemical reactions. The classification and specification of organism was based on the scheme presented in Bergey’s Manual of Systemic Bacteriology.

Out of 153 samples 113 samples were collected from high yielding Friesian crossbred cattle (40 from Bonoful dairy farm and 73 from local mini dairy farms) and 40 from Red Chittagong cattle (all from local mini dairy farms). The overall prevalence of subclinical mastitis was found 65.35% . In case of Red Chittagong cattle, the prevalence was 12.5%, while incase of Friesia crossbred cattle (Bonoful dairy farm), the prevalence was 80% and incase of Friesian crossbred cattle (local mini dairy farms), the prevalence was 86.30%.
