Genetic Conservation and Quality Seed Production of Indigenous Catfishes (heteropneusstes Fossllis and Clarias Batrachus) Through Cryogenic Gene Banking.

Abstract/ Executive Summary:

A series of experiments were conducted to develop and standardize the cryopreservation protocols of shing (Heteropneustes fosilis) and magur (Clarias batrachus). The activation of sperm of both shing and magur mixed with three extenders such as Alsever’s solution, urea egg-yolk and egg-yolk citrate was tested at different concentration of salt solution and was found that the motility of sperm decreased with the increase of Nacl concentration. The motility of sperm was ˃90% up to 0.3% Nacl but it became zero at beyond 0.8% Nacl solution. Toxicity of cryoprotectant to sperm was investigated at 5, 10 and 15% concentrations of cryoprotectant with an incubation period of 5 to 20 minutes. Sperm motility decreased with the increase of cryoprotectant concentration and incubation time for all three cryoprotectants (methanol, DMSO and ethanol). At 10 minutes incubation, motility of sperm was ˃60% at 5 and 10% of cryoprotectant but it has been decreased to below 30% at 15% of those tested cryoprotectants. Sperm of shing was preserved with four extenders (Alsever’s solution, Ginzburg Fish Ringer, egg-yolk citrate, urea egg-yolk) and three cryoprotectants (DMSO, methanol, ethanol). The equilibrium and post-thaw motility of sperm preserved with Alsever’s plus either DMSO, methanol and ethanol was between 80 to 85%, and 40 to 60% respectively. Sperm preserved with Ginzburg Fish Ringer plus DMSO and methanol produced ˃75% and ˃65% equilibrium and post-thaw motility respectively. Sperm preserved with egg-yolk citrate and urea egg-yolk demonstrated ˃85% equilibrium motility which reduced to 80% at post-thaw period. In case of magur, sperm preserved with three extenders such as Alsever’s solution, egg-yolk citrate and urea egg-yolk and two cryoprotectants DMSO and methanol. Alsever’s solution plus DMSO and methanol produced ˃85% equilibrium and ˃55% post-thaw motility while egg-yolk citrate plus DMSO and methanol produced ˃85% and ˃45% equilibrium and post-thaw motility respectively. Sperm preserved with urea egg-yolk plus DMSO and methanol have shown 85% equilibrium and ˃50% post-thaw motility. Two breeding trials of magur using cryopreserved sperm were conducted. Sperm preserved with Alsever’s + DMSO produced 10% fertilization and 5% hatching while 25% fertilization and 17% hatching were recorded from fresh sperm.

Achievement/Findings:
