Artificial Propagation and Rearing of an Indigenous Fish Species, Rani, Botia Dario

Abstract/ Executive Summary:

In order to get information on the reproductive activities of Botia Dario, a total of 110 adult fishes have collected from the different places of greater Mymensingh district, during January-February 2009. The collected live fishes have stocked in a pre-prepared pond of the field laboratory complex, Faculty of Fisheries, Bangladesh Agricultural University (BAU), Mymensingh. The average length and weight were 8.0 cm and 35g respectively. The male and female ratio was 1.2:0.8 respectively. A special feed enriched with protein and vitamin-E was formulated and supplied at @ 5% of body weight of the brood fishes, twice daily. During June-July, matured males and females were captured from the experimental pond. The brood fishes were brought at the hatchery of laboratory complex and released in the aquarium with water shower 6-8 hrs for adjust at the room temperature for over night. Two different types of methods (striping and natural) were applied for induced breeding in the captive condition but no positive result was observed in any attempt. Carp pituitary gland (cPG) extract (60mg and 20mg for female and male respectively) and ovapin-super (4ml and 2ml for female and male respectively) was applied for injection purpose in both the cases. Four matured females and eight matured males were used for each hormone. For striping method, the injected males and females were striped after 6 hours of decisive doses and the milt mixtures eggs were stocked metallic trays with water shower. Then the eggs were observed carefully up to 72 hours but no hatching was occurred in any cases.

Objectives:

1) To determine the effective dose of carp pituitary gland and HCG as the inducing agents to breed Botia dario; 2) To study the fertility rate, hatching rate and embryonic development process of this species; 3) To study the effects of different natural and artificial feeds on the growth and survival of larvae for rearing them up to the stockable size.

Methodology:

The brood fish of Botia dario will be collected from different places of Mymensingh district for breeding purpose. As soon as the fish reaches the ripe stage, they will be brought to the hatchery and injected with HCG and carp pituitary extract at different doses to find out the appropriate hormone dose for inducing ovulation. The eggs will be fertilized with milt from male rani and placed to the hatching jar for incubation. The hatching jar will be supplied with aeration to ensure sufficient oxygen to the developing embryos. Different methods and techniques will be applied to increase the hatching success of the fertilized eggs on trial and error basis. Time and temperature will be recorded. Embryonic developmental progress will be studied as per method followed by Binoy Chakraborty, 2004. The spawn will be transferred to the well prepared nursery pond to observe their survival and growth rate with different ingredients rich supplemental diet. After a period of two months, the fingerlings will be released in grow out ponds. Monthly growth rate, length weight relationship, fecundity, average daily weight gain, specific growth rate, water quality parameters etc. will also to be studied.

Achievement/Findings:

In case of natural method, carp pituitary gland (cPG) extract and Ovapin-super were applied as same administration like striping method. Four matured females and eight matured males were used for each case. The injected males and females stocked at hapas in the pond environment after the decisive doses and different types of water hyacinth were used in the hapas for created the sheltered the fishes and were observed carefully up to 72 hours after 18-20 hrs the belly was became gradually harder and lastly damaged eggs were released with striping in any cases.
