Rice (Oryza sativa) plants accquired excess photosynthates in the form of non-structural carbohydrates (NSCs) in their stems on the eve of heading which successively translocate to grain during grain-filling. Despite keen interest in stem and grain NSC for rice improvement, the dynamics of NSC accumulation, re-translocation, and re-accumulation that have genetic potential not yet been perfectly investigated. Here we conducted three experiments, greenhouse and field to lay the groundwork for large-scale diversity studies on rice stem and grain NSC including some agronomic traits. Both greenhouse and field experiments were conducted under direct-seeded rice (DSR) system with flooding at the International Rice Research Institute, Philippines, using elite lines incorporating AG1, AG2 and AG1-AG2 into the popular varieties PSBRc82 and Ciherang-Sub1 along with the donors Kho Hlan On (AG1) and Ma-Zhan Red (AG2). In germinating seedlings soluble sugars increased, while starch concentration decreased gradually under flooding especially in the tolerant checks and AG1-AG2 introgression lines. Stem soluble sugarb accumulation started to increase from vegetative stage and peaked at panicle initiation stage then gradually decreased towards maturity stage. But Sub1-AG lines had higher sugar and starch concentration in different growth stages than other genotypes in both seasons.

To lay the groundwork for large-scale diversity studies on rice stem and grain NSC including some agronomic traits. Both greenhouse and field experiments were conducted under direct-seeded rice (DSR) system with flooding.

These studies were performed in the green house and research farm of International Rice Research Institute (IRRI), Philippines (14°08′ N, 121°15′ E, 23 m altitude from sea level) during the 2016 wet season (from June to November) and the 2017 dry season (from January to May). One Greenhouse experiment was conducted for nonstructural carbohydrates studies and two field experiments were done to find agronomic performance as well as nonstructural carbohydrates dynamics of tested genotypes. In greenhouse experiment and first field experiment, ten rice genotypes with anaerobic germination (AG) potential i.e., AG1, AG2 and AG1-AG2 introgression lines PSB Rc82-AG1, PSB Rc82-AG2, PSB Rc82-AG1-AG2, Ciherang-Sub1-AG1, Ciherang-Sub1-AG2, Ciherang-Sub1-AG1-AG2, along with their donors Kho Hlan On (AG1), Ma-Zhan Red (AG2) and intolerant recipients PSB Rc82 and Ciherang-Sub1 were used.
The greenhouse experiment was conducted by using seedling trays with grids of 34 rows, each row comprising 17 cells of 1 cm × 1 cm × 2.5 cm filled with finely ground field soil. Dry seeds were placed in seedling tray, which consisted of 578 (34 × 17) cells half-filled with sterilized garden dry soil, then one seed was placed and covered with 1 cm of fine soil. With 51 seeds for each entry the trays were placed in concrete tank and was submerged under 10 cm water depth, likewise another set of trays was maintained under control (saturated) conditions in an adjacent area up to 21 days after sowing (Ismail et al, 2009; Ella et al, 2010; Septiningsih et al, 2013) in a randomized complete block design with three replications. First field experiment was laid out in a randomized block design with three replications (n = 3) having experimental unit size 12 m2 (4 m × 3 m) using aforesaid ten rice genotypes.
