Genetic Mapping for Quantitative Trait Loci for Maturity of Mungbean (vigna Radiata L)

Abstract/ Executive Summary:

Mungbean (Vigna radiata (L) Wilczek) varieties with synchronous maturity is generally preferred for cultivation. In this study, we report quantitative trait locus (QTL) mapping in Mungbean using F2 population of 142 lines derived from a cross between KPS-I (75-80 days) and BARI Mung 6 (55-60 days). F_2:3 lines grown under field condition at Gazipur 2016 and 2017. Single marker analysis suggested at least five loci controlling maturity. Composite interval mapping consistently identified four QTLs, qMAT1,qMAT2, qMAT3, qMAT6 and qMAT8, on linkage groups 1, 2, 3,6 and 8 for both years 2016 and 2017. These QTLs accounted for 7.54 to 18.83% variation depending on different linkage group.

Objectives:

To identify the quantitative trait locus (QTL) mapping in Mungbean using F₂ population of 142 lines derived from a cross between KPS-I (75-80 days) and BARI Mung 6 (55-60 days).

Methodology:

The F_2:3 progenies and parents were evaluated for synchronous maturity at of PRSS, Gazipur for two years i.e. 2016 and 2017. 142 F_2:3lines were sown together with their parents in randomized complete block design (RCBD) with two replications. Each entry was sown in two rows with 2 m long. Date of sowing was 19/3/2017. Row to row distance was 40 cm with spacing between adjacent plants of 10 cm. Fertilizers were applied at 40 kg/ha urea, 90 kg/ha triple super phosphate, 60 kg/ha muriate of potash and 40 kg/ha gypsum prior to soil preparation following recommendation guide of BARI for Mungbean. Weeds were controlled manually. Harvesting was done by hand. The average data of 80% pod maturity in each entry were used for statistical and QTL analysis in each of the experiments. Statistical analysis: Analysis of variance (ANOVA) was carried out for the disease score data from individual experiment and combined data using the Win QTL Cartpgrapher and JoinMap 4.0 (Van Ooijen 2006) software. The linkage groups were calculated with the minimum logarithm of the odds (LOD score) at 3.0, the maximum recombination frequency (r) at 0.5 and Kosambi mapping function (Kosambi 1944). Marker scores were subjected to goodness of fit test using chi-square analysis (1:2:1 and 1:1 ratios in the F_2:3populations).

Marker(s) associates with synchronous maturity were preliminarily determined by single marker analysis at P = 0.001. At Gazipur, 21 markers from 5 LGs (1, 2, 3, 6 and 8) associated with synchronous maturity for both the year. Composite interval mapping (CIM) employed to locate the QTL onto linkage map. Significant thresholds for QTLs were determined by permutation test. CIM identified four QTLs for maturity on different linkage groups. The QTLs were each on LG1, LG2, LG3, LG6 and LG8 and were designated as qMAT1,qMAT2, qMAT3, qMAT6 and qMAT8 respectively. Among these QTLs qMAT2 had the largest effect on maturity. QTLs qMAT1had the lowest effect on explained phenotypic variation.
