Screening of Mungbean Germplasm Under Nacl Salinity. Season 2017-2018

Abstract/ Executive Summary:

Study the effect of salinity on germination, root and shoot growth of Mungbean (Vigna radiata L.) at four salinity levels e.g. 0, 2, 4 and 6 ds/m of NaCl concentrations was conducted at PRSS GAzipur. Salinity affects imbibition, germination and root elongation. Highest germination % was observed in control treatment of all the varieties. Root growth was significantly reduced with higher NaCl concentrations, and genotypes ML 1299 showed better performances than other varieties. All the varieties showed similar performances at higher NaCl concentration considering yield contributing character.

Objectives:

To study the effects of salinity on germination, growth Vignaradiata varieties.

Methodology:

The experiment was conducted in 2018 at laboratory of PRSS, BARI, Gazipur. Seed of three Mungbean genotypes (VC 6368, VC 6372 and ML 1299) were used. Seeds were sown in Petridis with blotting paper soaked with different concentration of salt solution. Four different concentrations 0, 1, 2, 3 and 4ds/M conc. salt solution were used. For this purpose the salt solution different amount of NaCl was dissolved in 1 L water as per described by R Munns and R A Richards (1998). There was sufficient amount of solution used to soak the each petridis. The each was watered as and when required and care was taken not to leach the salt by excess watering. There were 3 replications for each treatment and the experiment was arranged in a complete randomized design. Electrical conductivity (ECe) of different treatments was measured by using a conductivity meter (Field scout soil and water EC meter). Germination percentage, root & shoot length were recorded from the treatment of range salinity (0, 1, 2, 3 and 4 ds/M NaCl conc.). Data on root and shoot development was recorded after three days of seed placement on petridish.

Achievement/Findings:

The effect of NaCl concentration on root growth of Mungbean varieties at different dates presented. The response of Mungbean at different dates showed remarkable variation among the varieties. During first developmental stage, salinity caused considerable delay and reduction in seed germination. Among the varieties VC 6372 did not show any remarkable variation among the different dates. VC 6368 and ML 1299 showed significant difference at different dates. The highest germination (%) was observed at control treatment and which was followed by 1, 2 3 and 4 ds/M. Among three genotypes ML 1299 showed highest germination % considering all treatments. Higher conc inhibits germination of Mungbean. Wests and Francios (2004) reported germination significantly reduced when electrical conductivity increased in case of cowpea. In case of VC 6368, at 4 days and 5 days after germination root growth showed significance variation. The highest root growth was found at control treatment which was followed by the higher concentration of NaCl treatment. Considering the root growth of three genotypes ML 1299 was found better than the other two genotypes. Balasubramanian and Sinha (2006) reported that root growth was more sensitive to increase in salt stress both cowpea and Mungbean. But in shoot development VC 6368 performed better than other genotypes. All the varieties showed significant difference at later stage for shoot development. The electrical conductivities were also measure in petdrishis and was 1.05, 2.02, and 3.20 dSm^-1 respectively after five days of placement. Apart from this salinity stress increasing salinity is also accompanied by significant reductions in shoot weight, plant height and root length (Parida and Das 2005; Hajier et al. 2006).The experiment not yet completed, It will continue in next year for physiological and yield related trait studies.
