Screening of Hybrid Tomato Genotypes Against Salinity During Germination and Early Seedling Growth Stage (2017 – 2018)

Abstract/ Executive Summary:

A lab experiment was carried out at the Plant Physiology Laboratory of HRC, BARI, Gazipur during December 05, 2017to December 30, 2017to find out the salinity tolerant tomato genotypes during germination and early seedling growth stages. The response of six hybrid tomato genotypes viz. BARI Hybrid Tomato 8 (V₁), BARI Hybrid Tomato 9 (V₂), BARI Hybrid Tomato 4 (V₃), BARI Hybrid Tomato 5(V₄), BARI Hybrid Tomato 7(V₅), Hybrid Tomato (Unnyan) (V₇), Hybrid Tomato (Mintu) (V₆), Hybrid Tomato (Comet) (V₈) against four levels of salinity viz. 0 dS/m (S₀), 4 dS/m (S₁), 8 dS/m (S₂) and 12 dS/m (S₃) were studied in this experiment. The germination rate and seed vigour index along with other attributes were reduced with increasing the salinity levels. Compared with 8 tomato genotypes, BARI Hybrid Tomato 8 (V₁) performed relatively better at 12 dS/m with respect to germination and early seedling stage that was followed by BARI Hybrid Tomato 9 (V₂), BARI Hybrid Tomato 4 (V₃) and BARI Hybrid Tomato 5 (V₄). The performance of Hybrid Tomato (Comet) (V₈) was poor compared to other varieties. Therefore, it can be concluded that BARI Hybrid Tomato 8 (V₁), BARI Hybrid Tomato 9 (V₂), BARI Hybrid Tomato 4 (V₃) and BARI Hybrid Tomato 5 (V₄) may salt tolerant at germination and early seedling growth stage. The performances of all genotypes were found inferior at 8 and 12 dS/m salinity levels but survived up to 12 days after germination.

Objectives:

To identify the tomato genotypes under different salinity stress with respect to germination and early seedling stage.

Methodology:

The experiment was conducted at the laboratory of Plant Physiology Section, Horticulture Research Centre, BARI during December 05, 2017 to December 30, 2017. The experiment consisted of Eight hybrid tomato genotypes viz. BARI Hybrid Tomato 4 (V₁) , BARI Hybrid Tomato 5 (V₂), BARI Hybrid Tomato 7 (V₃), BARI Hybrid Tomato 9 (V₄), BARI Hybrid Tomato 8 (V₅), Hybrid Tomato (Comet) (V₆), , Hybrid Tomato (Mintu) (V₇), Hybrid Tomato (Unnyan) (V₈) considered as factor A and four levels of salinity viz. 0 dS/m (S₀), 4dS/m(S₁), 8 dS/m(S₂), 12dS/m(S₃) considered as factor B. The seeds were collected from Horticulture Research Centre, BARI and Plant Genetics Resource Center (PGRC), BARI, and local market were used in the study. The NaCl concentrations were measured by EC Meter. Required number of petridishes each with a diameter of 11 cm were used in the experiment and arranged in a completely randomized design (CRD) moistened with respective treatment in three replications. Three whatman filter paper no.1 were used in each petri dish for retaining required amount of water for imbibition of seeds. Twenty healthy and equal sized seeds of each variety were selected and placed on sterilized petridish lined with filter paper. The pertidishes with seeds of the selected varieties of tomato were kept in normal room temperature for germination. The seeds were sterilized by soaking in a 5% solution of sodium hypochlorite for 5 min and placed on the petridish lined with filter paper. The germination count was taken after 72 hours of placing seeds. The 5 ml of distilled water (control) and 5 ml solution of salinity were added separately in the petri dishes. The petridishes were covered to prevent the loss of moisture by evaporation. Number of seeds germinated was counted daily and data recording were continued up to 12 days. After 12 days 10 seedlings (shoot + root) were weighed by the electrical balance and this weight was considered as fresh weight of seedling, and these seedlings were kept in an oven at 72^Oc for 1 weeks for recording dry weight of seedling. A seed was considered to have germinated when both the plumule and the radicle emerged > 0.5cm. After 12 days, the shoot and the root length of ten randomly selected seedlings from each replicate were measured. The analyses were done following the software STATISTICA, Version 5 (Statsoft France, 1997). The significance of the difference among the means was evaluated by the Least Significant Difference Test (LSD) at 5% level of probability.

Achievement/Findings:

Compared with 8 tomato genotypes, it can be concluded that BARI Hybrid Tomato-8 (V₁), BARI Hybrid Tomato-9 (V₂), BARI Hybrid Tomato-4 (V₃) and BARI Hybrid Tomato -5 (V₄) performed better upto 8 dS/m salinity levels compared to other genotypes at germination and early seedling growth stage. However, further trial is needed for confirmation of the result including more genotypes as per collection.
