Study on the Disease Reaction of Guava Native Germplasm Against Nalanthamala Psidii a New Pathogen of Guava Wilt

Abstract/ Executive Summary:

An experiment was carried out during January to June, 2020 in pot house, Plant Pathology Division, BARI to evaluate the response of some native guava lines against wilt disease caused by Nalanthamala psidii under artificially inoculated condition. Ten native guava lines viz. LOG1, LOG2, LOG3, LPG1, LPG2, LMG1, LMG2, LMG3, TMG1, TMG2 and one check BARI peyara 2 were tested against the disease. Disease reaction was observed after inoculation of mycelial block in trunk of stem. Two lines LPG1 and LMG1 showed lower disease incidences 14.66 and 17.33%, respectively after six months of inoculation. The highest disease incidence 40% was observed the line TMG1.

Objectives:

To find out a resistant germplasm of guava against Nalanthamala psidii causing wilt disease.

Methodology:

Guava was collected from home garden and orchard as well as local market of Gazipur. Four or five number of fruits were collected from single guava tree or lot. Internal part of guava with seed was placed in small plastic container containing water and kept to rot for 5-10 days. Then seed was separated and air dried for 48 hours. Collected seeds were stored at room temperature in plastic zipper bag. In this experiment, NPB-001 isolate of N. psidii used as pathogen was collected from stock culture of Plant Pathology Division, BARI. The fungus was grown on PDA media at 25⁰C for 7 days. Three mycelial plugs of 5 mm diameter were inserted into a 2 cm² bark flap on the trunks, 20 mm above the soil line, of three-month-old seedlings of guava lines grown in pots. The wound was gently wrapped with parafilm. The control was inoculated by inserting PDA plugs only in the trunk. Plants were evaluated six months after inoculation, using a scale of 1–5 where 1 = four or less leaves turned purple, 2 = five or more leaves turned purple, 3 = quarter of plant wilted (>8 leaves turned purple), 4 = half of plant wilted and 5 = dead plants. The disease index was calculated as (sum of all numerical ratings/total number of plants) × (100/maximum disease category)

Achievement/Findings:

LPG1 and LMG1 showed low levels of susceptibility to wilt disease causing pathogen N. psidii. These two lines will be evaluated under more disease pressure in next two years. More lines will be collected from throughout the country and evaluated next year.
