Collection, Isolation and Identification of Xanthomonas Campestris Pv. Campestris from Cabbage and Cauliflower

Abstract/ Executive Summary:

Black rot caused by Xanthomonas campestris pv. campestris is the major constraint for the cultivation of cabbage and cauliflower which is one of the most popular winter vegetables grown in Bangladesh. Twenty-two disease infected leaf samples were collected from different cabbage and cauliflower growing areas of Gazipur district during 2018-2019 cropping season. The pathogens were then isolated, purified and preserved following the proper procedures. The bacterium was identified through morphological characteristics on different solid media. With regard to morphological variations, the isolates varied in their morphology on different solid media used for screening.

Objectives:

The study was taken to identify and characterize the bacteria Xanthomonas campestris pv. campestris

Methodology:

Collection of black rot infected leaf samples The leaf samples were collected from different cabbage and cauliflower growing areas of Gazipur district during the 2018-2019 cropping season. From each field, a leaf with typical black rot symptoms (V-shaped lesions) was collected from cabbage and cauliflower plants. A total of 22 black rot samples were collected from a different area of which 12 from cabbage and 10 from cauliflower. Samples were dried between sheets of paper at room temperature before isolation of the causal agent.

Isolation and preservation From each of the 22 black rot bacterial samples collected, 0.5 to 1 cm² of leaf tissue was excised with a sterile scalpel from the margin of a lesion and placed in a drop of sterile 0.85% saline for 5 min. A loopful of the saline suspension was streaked on YPSA plates. The plates were then incubated at 28 ^OC for 48-72 hours according to Schaad and Stall (1988). Bacterial colonies from each plate were further sub-cultured and pure bacterial colonies were transferred to YPSA plates incubated at 28 ^OC for 48-72 hours and preserved at 4 ^OC for further work.

Cultural characteristics of different isolates of Xanthomonascampestris pv. campestris from cabbage and cauliflower on different solid media The physiological characters of the isolate of the bacterium were studied for colony morphology on YDCA, YNA, and TSA medium. The test was conducted as per the methods described by Bradbury (1970) and Schaad (1988).

Achievement/Findings:

From the study, it was observed that there were considerable variations in colony characters on different solid media, which gave clue to the presence of variations within Xanthomonas campestris pv. campestris populations in Bangladesh. This is valuable information to further study like other morphological, physiological, biochemical, and molecular characteristics which will be helpful to find out the cause for the variations within the isolates and for devising various control measures.
