In-vitro Efficacy of Anitbiotics Against Bacteral Leaf Spot of Tomato Caused by Xanthomonas Campestris Pv. Vesicatoria

Abstract/ Executive Summary:

In February 2019, suspected Xanthomonas campestris pv. vesicatoria infected tomato leaf samples were collected from Jamalpur district. After isolation on a semi-selective CKTM medium,the isolate was confirmed through biochemical tests. In the sensitivity assessment of the isolate against different concentrations of commonly used chemical components namely crosin-Ag 10sp (streptomycin sulphate 9%+tetracycline hydrochloride 1%) and sunvit (copper-oxychloride 50%) by McDonald Bangladesh PVT. Limited, the isolate was highly resistant as it grew on the medium with 200 µg/L of chemicals.

Objectives:

Determination of optimum concentration of bactericides against the bacteria is very necessary and it is a need of time.

Methodology:

In February 2019, bacterial leaf spot infected tomato leaf samples were collected from Jamalpur district. Samples were transported to the laboratory of the Plant pathology Division, BARI and culturing were initiated within the days of collection. The bacterial suspension was made by macerating the spotted tissue with green tissue in sterilized distilled water (5mL) and the extract was streaked on selective media CKTM. After that, the pure yellow color colonies were streaked on Calcium carbonate agar (YDC) media twice and were placed at 28°C for 3 days. The purified isolated colonies were confirmed through biochemical tests followed by Schaad et al. (2001): gram reaction (3%KOH), oxidase disk, fluorescent on Pseudomonas semi-selective PF media, ability to grow on YDC medium. For the sensitivity assessment, the isolate was streaked on Luria-Bertani agar medium (LBA) for 48 hours at 28°C and adjusted to an optical density of 0.2 (OD₆₀₀). Bacterial concentration was confirmed by dilution plating (10⁸CFU/mL). Fifty microlitres of the suspension were transferred to LBA medium containing 25µg/L, 50µg/L, 100µg/L, 200µg/L of Crosin-Ag 10sp (streptomycin sulphate 9%+tetracycline hydrochloride 1%) and sunvit (copper-oxychloride 50%) by McDonald Bangladesh PVT. Limited. Copper fungicides were included since the extensive use of copper to manage the bacterial disease of crops for more than 100 years.

Achievement/Findings:

The results for the biochemical tests of the isolate are provided in Table1. The isolate was gram-negative, oxidase negative did not give fluorescent on PF medium, mucoid growth on semi-selective YDC medium. After incubation of 72 hours at 28°C for sensitivity assessment, four different categories were decided to evaluate the resistance of the isolate against different ppm of copper and streptomycin; sensitive, S= no growth at 25µg/L; moderate resistant, MR= growth at 50µg/L; resistant, R= growth at 100µg/L and highly resistant, HR= growth at 200µg/L. From this in-vitro assessment, it was found that the bacterial leaf spot causing tomato isolate is highly resistant to both streptomycin and copper while the plates which were non-amended LBA for comparisons to treated one remained full growth of bacterial colonies.
