Validation Trial of Bacillus Oryzicola Yc7007 for Controlling Bacterial Wilt in Eggplant (2018 - 2019)

Abstract/ Executive Summary:

Bacterial wilting for especially eggplant is a serious threat over the world. Still, there is no successful remedy to control the wilt in eggplant. However, from the last decade, Bacillus species and their products have been used in agriculture with many aspects over the world. This approach is rudimentary in our country. The activity using the endophytic novel Bacillus oryzicola YC7007 to control wilt in eggplant is the first study in our country to our knowledge. The in vitro antagonistic study had been conducted successfully at the Regional Agricultural Research Station, Hathazari, Chattogram to control the bacterial pathogen Ralstonia solanacearum pv. brinjal RARS Hat3 by the strain YC7007 since 2016. Strain YC7007 did quorum having with the 2.0 × 10⁷ CFU/ml inocula that suppressed the wilt and promoted the plant growth compared with control. Out of six varieties (BARI Bt Begun 2, BARI Bt Begun 3, BARI Bt Begun 4, BARI Begun 4, BARI Begun 5 and BARI Begun 6), BARI Begun 6, BARI Bt Begun 4 and BARI Bt Begun 3 showed the disease reduction followed by BARI Begun 5, BARI Bt Begun 2 and BARI Begun 4. Strain YC7007 (2.0 × 10⁷ CFU/ml) revealed significantly (P < 0.05) lower disease severity by 0.33 ± 0.3, than the control 1.0 ± 0.5 in the BARI Bt begun 4 at 5 Month after Transplanting (MAT). However, strain YC7007 (2.0 × 10⁷ CFU/ml) revealed significantly (P < 0.05) lower disease severity by 1.0 ± 0.5, than the control 1.67 ± 0.6 at BARI Bt begun 3at 5MAT. The strain showed significantly consistent disease suppression 100-67% in the BARI Bt begun 4 and 100-41% at BARI Bt begun 3 during 4 MAT to 5 MAT respectively to wilting compared to control.

Objectives:

To evaluate the efficacy of identified novel stains this experiment has been undertaken against wilting of BARI Bt begun 2, BARI Bt begun 3, BARI Bt Begun 4 and with their corresponding non-Bt brinjal viz. BARI Begun 4, BARI Begun 4 and BARI Begun 6 respectively.

Methodology:

The experiment was conducted at Plant Pathology laboratory, Regional Agricultural Research Station, Hathazari, Chattogram from 2016-2019 to find out the efficacy of strain YC7007 and to confirm its validation test against wilt of eggplant. Six varieties of eggplant viz. BARI Bt Begun 2, BARI Bt Begun 3, BARI Bt Begun 4, BARI Begun 4, BARI Begun 5 and BARI Begun 6 were subjected to conduct the experiment. Pinprick, stem clipping and root clipping bioassay were conducted to challenge the pathogen from 2016 to 2017 in vitro for the screening test. In the field trial, direct artificial inoculation has been omitted in the current year 2018-2019. The bacterial suspension was made with a buffer of 10 mM MgSO_4.Two times drenching by 300 ml bacterial suspension for each plant was done and one time spraying with the bacterial suspension was made by the 2.0 × 10⁷ CFU/ml inocula till to droplet of suspension from the leaves. Fertilizers and irrigation were maintained accordingly in homogenous conditions to all (Azad et al., 2017). The data of bacterial wilt of eggplants were generated by the following rating scales. 0 - Highly Resistant (HR) with no wilt symptom; 1 -Resistant (R), with 1 - 10% wilted plants; 2 – Moderately Resistant (MR) with 11 -20% wilted plants; 3 – Moderately Susceptible (MS), with 21-30% wilted plants; 4 – Susceptible (S) with 31- 40% wilted plants, and, 5 - Highly Susceptible (HS) with > 40% wilted plants. Data were analyzed by the HSD Tukey at 1-5% level having three replications.

Achievement/Findings:

The inocula for quorum sensing by the novel strain YC70007 is 2 × 10⁷CFU/ml. Strain YC7007 is a strong antagonistic endophytic bacterium against wilting of BARI Bt Begun 4 and BARI Bt Begun 3. The strain showed significantly consistent disease suppression 100-67% in the BARI Bt begun 4 and 100-41% at BARI Bt begun 3 during 4 MAT to 5 MAT respectively, against wilt in Bt Brinjal compared to control.
