Survey and Immuno-nucleobased Based Diagnosis of Cucurbit Infecting Viruses in Northern Part of Bangladesh

Abstract/ Executive Summary:

A survey was conducted in Rajshahi, Natore and Joydebpur to assess viral diseases of cucumber, Pumpkin, Bottle gourd and Bitter gourd. In the surveyed area different types of viral diseases are prevailing to infect cucurbits crop including mosaic, yellowing, mottling, leaf curling and bud necrosis with the percent incidence of 30-80%. The DAC-ELISA coupled with PCR and sequencing revealed that the identified virus was Papaya ringspot virus-P/W, Cucumber mosaic virus and Geminivirus. The other viruses of these locations are now in progress.

Objectives:

Under this proposed project, the clear picture of emerging cucurbits infecting viruses and their vectors may come out which will directly help to develop a durable integrated control strategy against the viruses

Methodology:

The survey has been conducted in three different cucurbits growing upazilla namely Bagmara, Mohanpur & Godagari at Rajshahi, Districts, Gorudaspur upazilla of Natore and Experimental field of Plant Pathology, Division were surveyed to assess the viral diseases of cucumber, pumpkin, bottle gourd and bitter gourd. The virus diseases like samples were collected and brought to the Plant Virology Lab, PPD, BARI for further analysis. For Serological detection, DAC-ELISA has been performed. Plant genomic DNA has been isolated using C-TAB method and RNA isolated using SV total RNA isolation system (Promega). The RNA viruses RT-PCR done by specific primers, retrieving sequences from NCBI and aligned them using BioEdit software version 5.

Achievement/Findings:

From the results of DAC-ELISA and PCR followed by sequencing PRSV-P/W, CMV has been diagnosis from Cucumber, Pumpkin, bottle gourd. The leaf curl samples of cucumber Gemnivirus have been amplified using Geminivirus universal primers and the identification of the Geminivirus species is now in progress. The genetic diversity of cucurbits infecting viruses and the identification of new viruses are undergoing by using molecular tools.
