Isolation and Identification of Different Diseases in Pomegranate (punica Granatum) (2016 - 2017)

Abstract/ Executive Summary:

An experiment was carried out to isolate and identify fungal and bacterial diseases in pomegranate in Rajshahi region during 2016-17 and found two new diseases. Wilt of pomegranate is an important fungal disease and its causal organism, Ceratocystis fimbriata were successfully identified on the basis of morphological characteristics of culture and conidial shape of the pathogen. Pathogenicity was confirmed following slandered procedure. Bacterial disease Xanthomonas axonopodis pv. punicae were isolated from infected leaves and fruit. Morphological and biochemical characteristics of bacteria were found after the completion of different tests. To the best of our knowledge, both are the first reports on pomegranate in Bangladesh.

Objectives:

The present study was undertaken to identify the prevailing fungal and bacterial diseases in pomegranate leaves and fruits

Methodology:

Different infected parts of pomegranate leaves and fruits showing typical symptoms were collected from different locations of Rajshahi District. Diseased root of the pomegranate tree was collected from the farmers’ field of Naogaon District. The fungus and bacteria were isolated from the infected parts of pomegranate following standard phytopathological procedures.

Isolation of Bacteria

We collected infected plant leaves and fruits to isolate the bacterium. All the samples were carried to the laboratory in sterile polythene bags. The fruit and leaf samples were washed thoroughly with tap water and allowed to dry. The samples were surface sterilized with 0.1% mercuric chloride (HgCl₂) solution for one minute and washed three times serially in sterile distilled water to remove the traces of mercuric chloride. The infected portion was cut into small pieces and suspended in 10 mL of sterilized distilled water and squeezed gently with a sterilized scalpel. When the water became slightly turbid due to bacterial cells, then a drop of the suspension was spread on the sterilized nutrient agar Petri plates. The inoculated plates were incubated at 30°C for 48 hours. The plates were observed for the development of well separated, typical, yellow colored bacterial colonies resembling Xanthomonas sp. The two bacterial isolates obtained upon isolation from the diseased samples were stored at -20°C in glycerol stocks.

Morphological and biochemical characterization of the isolates for identification

The colony growth and morphological characteristics of the pathogen such as cell shape, colony color, Gram stain reaction characters were studied. The biochemical characters such as catalase, oxidase, MR test and Indole production test of the isolates were studied. The isolates were identified by comparing the characteristics with their description reported in Bergey’s Manual of Systematic Bacteriology.
