Biological Control of Wilting by Endophytic Bacteria Bacillus Sp. Yc7007 in Eggplant (2016 - 2017)

Abstract/ Executive Summary:

The in vitro experiment was conducted at the Regional Agricultural Research Station, Hathazari, Chittagong during the period 2016-2017 to control the bacterial wilt caused by Ralstonia solanacearum pv. brinjal RARS Hat3 in the BARI Bt begun 3. Its pathogenicity test by the Koch’s Postulates and finally its suppressive bioassay by the novel species Bacillus sp. YC7007 was conducted. Strain YC7007 (2.0 × 10⁷ CFU/ml) revealed significantly (P < 0.01) lower disease severity by 3.27. ± 0.51, than the control 4.67 ± 0.16 by the artificial inoculation in the BARI Bt begun 3 at autoclaved soil condition. However, at the natural condition, strain YC7007 (2.0 × 10⁷ CFU/ml) revealed significantly (P < 0.01) lower disease severity by 3.10 ± 0.32, than the control 4.50 ± 0.17 at that same cultivar. On the contrary, Strain was evaluated in the BARI Begun 8 that is naturally resistant to wilting. The strain showed significantly consistent disease suppression to wilting compared to control.

Objectives:

To control biologically the wilting of BARI Bt brinjal 3

Methodology:

The laboratory-based experiment was conducted at the Regional Agricultural Research Station, Hathazari, Chittagong from 2016-2017 to find out the efficacy of strain YC700 and to confirm the pathogenicity by the new pathovars Ralstonia solanacearum pv. brinjal RARS Hat3. Two cultivars: BARI Bt begun 3 and BARI Begun 8 were subjected to conduct the experiment. Pinprick bioassay and infiltration were conducted to challenge the pathogen. R. solanacearum pv. brinjal RARS Hat 3 (1 x 10⁷ CFU/ml) and was infiltrated by needleless syringe at the abaxial surface of the leaves that was prepared by the 10 mM MgCl₂, 1 cc was infiltrated for the artificial inoculation. The pathogen was inoculated by spraying bioassay. Disease data were maintained by the following rating scales. 0 - Highly Resistant (HR) with no wilt symptom; 1 -Resistant (R), with 1 - 10% wilted plants; 2 – Moderately Resistant (MR) with 11 -20% wilted plants; 3 – Moderately Susceptible (MS), with 21-30% wilted plants; 4 – Susceptible (S) with 31- 40% wilted plants, and, 5 - Highly Susceptible (HS) with > 40% wilted plants. Data were analyzed by the student’s t-test and HSD Tukey at 1% level having three replications.

Bacillus sp. YC7007 controlled the bacterial wilting of BARI Bt begun 3 in vitro test by doing the artificial inoculation bioassay and it controlled 30 % disease reduction compared with control; however, YC7007 controlled 57% wilting in the BARI Begun 8 compared with control by the artificial inoculation by the pathogen
