Effect of Bacillus Oryzicola Yc7007 to Regenerate Grating Projection Overcoming Nursery Diseases (2016 - 2017)

Abstract/ Executive Summary:

The in vitro experiment was conducted at the Regional Agricultural Research Station, Hathazari, Chittagong during the period 2016-2017 to get successful grafting without the infection of nursery diseases. Novel species Bacillus sp. YC7007 was subjected to regenerate the reunion of scion and rootstocks. Strain YC7007 (2.0 × 10⁷ CFU/ml) revealed significantly (P < 0.01) good performance in the different plant growth-promoting parameters viz. number of leaves (8.95 ± 0.27), length of scion (15.8 ± 0.39 cm) and fresh weight (1.75 ± 0.05 gm / scion) followed by control number of leaves (5.65 ± 0.17), length of scion (9.2 ± 0.20 cm) and fresh weight (1.14 ± 0.02 gm / scion). YC7007 showed consistent grafting success 91-95% followed by control at 73-77.34% in the rough lemon and Kalamanshi rootstock respectively. Strain YC7007 showed a good grafting projection almost free nursery diseases. In the Rough lemon root-stock, Plant growth promotion (PGP) was highly expressed compared with the Kalamanshi, however numerical disease severity was higher in the rough lemon compared with Kalamanshi. About 88-90% scab fungal disease, 49-55% bacterial canker, and 100% dieback of Malta were controlled by the bacterial treatment.

Objectives:

To observe the bacterial interaction for making the good grafting projection.

Methodology:

The laboratory-based experiment was conducted at the Regional Agricultural Research Station, Hathazari, Chittagong from 2016-2017 to get a good successful grafting projection. Strain YC7007 (2 .0 × 10⁷ CFU/ml) mixed with 10 mM MgSO₄ inoculated at the part of scion cut and rootstock cut and then attachment by the veneer grafting was conducted. Twenty graftings were subjected under this bioassay under the individual treatment. After 20 DAI of YC7007, 1^st data were recorded. Data were analyzed by the student’s t-test at a 1% level.
