Collection and Assessment of Arbuscular Mycorrhizal Fungi in Different Field Crops (2016-2017)

Abstract/ Executive Summary:

A total of 24 crop samples from 12 crops covering 2 AEZs (AEZ-11 and 18) were collected and assessed during 2016-2017 for determining root colonization (%) and spore population (100g^-1 soil) of AM fungi. Gardenpea and potato showed the highest root colonization (30.0%) in AEZ-11 (Jessore) and motor (30.0%) in AEZ-11 (Ishurdi). But the highest number of spores (188.0 100g^-1 soil) was found with soybean in AEZ-18 (Rahmatpur).

Objectives:

To make a general survey of diversity in mycorrhizal association with agricultural crops and spore population in rhizosphere soils in different Agro Ecological Zones of Bangladesh.
To understand the dynamics of AM fungal root colonization and spore population for preparing a suitable arbuscular mycorrhizal inoculum for different crops.

Methodology:

Roots and rhizosphere soil samples were collected from the 2 AEZs namely 11 and 18 during December to February (2016-2017) when abundant plants were available in the fields. Five plants for each species were sampled. Plant roots were dug out, washed thoroughly with water to remove the adhering soil particles and then cut into 1 cm long segments. The root samples were then preserved in screw cap test tubes with 50% ethanol for future use. Soil samples were collected up to a depth of 0-15 cm with roots (3 samples/crop). Then the collected samples were air-dried, packed in airtight polyethylene bags and stored at 4^°C for assessing spore density. The percentage of AM infection was estimated by root slide technique (Read et al., 1976). Assessment of spore population was done by following the Wet Sieving and Decanting Method (Gerdemann and Nicolson, 1963).

Achievement/Findings:

Grasspea from two, lentil from three, chickpea from one, mungbean from one, gardenpea from four, groundnut from five, soybean from two, frenchbean from two and motor from one locations. These collected nodules were crushed, isolated and screened in the microbiology laboratory. Screening of some strains are going on.More crops covering different AEZs should be included in next year study. This programme will be continued in the next year.
