Papaya ringspot virus- W (PRSV-W) is one of the most serious virus caused disease in the pumpkin which affects leaves and also fruits in particular. A study was undertaken to develop a Papaya ringspot disease resistant hybrid in pumpkin at the Dept. of Plant Pathology during 2012/13. As a first step, twenty-nine germplasm lines maintained in the Dept. of Plant Pathology were evaluated for disease resistant. After seeing the disease response, germplasm lines were graded and hand pollination was done to get resistant F1 progeny. Then, some selected lines were used both as male (testers) and lines as female parents and 13 hybrids were obtained. All these hybrids were tested for the performance of yield, quality and disease resistance. An experiment was conducted of 13 hybrids and their 13 parents at the experimental field of Bangabandhu Sheikh Mujibur Rahman Agricultural University (BSMRAU), Salna, Gazipur, Bangladesh to assess their response to PRSV-W disease under inoculated PRSV-W condition during November 2012 to May 2013. The mean performance of growth, yield and yield contributing character of F1 progeny compared to parents in all cases showed better performances which was positive to select resistant hybrids. Of this population Hybrid 1, Hybrid 2 and Hybrid 11 exhibited increased resistance (0.0% disease incidence) exceeding their parental lines. Disease incidence was the parameters used to determine the resistance of population. The test genotypes varied significantly for their response to disease incidence (P = 0.000). Three hybrids (Hybrid 1, Hybrid 2 and Hybrid 11) for disease incidence demonstrated higher level of resistance. These entries can be released as Hybrid variety for PRSV-W resistance. Based on our results and previous literatures, disease incidence is an appropriate parameter for assessing the resistance of the genotypes to PRSV-W. Disease incidence inversely affects the plant growth and yield parameter as significant negative correlation (P < 0.05) exists between them. Significant relationship between disease incidence of the genotypes and plant growth suggested observed resistance of the genotypes was genetically controlled.

i.) To evaluate the selected F1 progeny and their parents of pumpkin under inoculated condition against PRSV-W disease. ii.) To compare disease incidence with growth and yield potential in selected F1 progeny and their parents in cross combination.

Selection of parental material The parents for the current study were selected from previous evaluation which was carried out through a breeding program in the Plant Pathology Department, Faculty of Agriculture, BSMRAU, Salna, Gazipur. The breeding program was started since two years ago (2010 - 2011) through earlier work which was done by the1 st Author. Thirteen selected pumpkin F1 progeny and their thirteen parents were selected depending on the degrees of resistance to PRSV-W disease, yield (kg/plant) at the experimental field of BSMRAU, Salna, Gazipur, Bangladesh. These parents were chosen as resistant, moderately resistant and susceptible lines according the former evaluation. Thirteen F1 were obtained by controlled pollinations among the selected 13 lines in all combinations, except reciprocals. Seed sowing, virus inoculation and transplanting The experiment was carried out for the 13 F1 progeny and their 13 parents. Seeds were sown in polybag in November 2012. The seeds of selected pumpkin crosses (13) with their respective parents (13) were planted in polybag (40 cm diameter) two seeds per polybag. PRSVW isolate was obtained from previously preserved infected leaf sample. The isolate used in this experiment was as described by Baker et al. 1991. Leaf samples of PRSV-W infected pumpkin plants were collected from the previous experiment and the samples were preserved following the procedures as described by Bos (1987). Inoculum of PRSV-W was prepared by grinding the infected pumpkin leaves using mortar and pestle in 0.02 M phosphate buffer, pH 7.0. Leaf to buffer ratio was 1:10 (1 g infected leaf to 10 ml buffer). The sap obtained after passing through double ply cheese cloth was used as inoculum. Mechanical inoculation method using carborundum powder (800 meshes, Fisher Scientific, Fair Lawn, NJ) was followed (Daryono, 2006). Inoculated old seedlings were transplanted in the main field. Plant to plant and plot to plot distance was maintained 2.0 m x 1 m, respectively. There were three replications; each replication contained four plants in this experiment. Randomized complete block designs (RCBD) with three replicates were used. The F1 was subsequently selfed and backcrossed to their respective parental lines, in order to obtain F2 generations and BC1 for all the crosses. Analysis of data The data were analyzed statistically by using the analysis of variance (ANOVA) and MSTAT-C software for proper interpretation. The mean value was compared according to Duncan’s Multiple Range Test (DMRT) at 5% level of significance. Diagram and graphs were used to interpret the data as and when required.
