Chlorella ellipsoidea was cultured in three different concentrations viz., 4.8, 7.2 and 9.6 g/I of BLA with added 0.4 g PSP and 0.2 g urea and in BBM (control) with triplicate for a period of three months. Each trial was continued for a period of 14 days in the laboratory. The initeal cell density of C. ellipsoidea was 2.5x105 cells/ml which attained the maximum of 94.86x105 cells/ml in BBM followed by 90.89x105, 83.88x10 and 71.34x105- cells/ml in 7.2, 9.6 and 4.8 g/I concentration of BLA, respectively on 10th day 5 of culture. The initial chlorophyll a content of inoculated C. ellipsoidea was 0.10 mg/I which attained the highest content 4.82 rng/I in BBM followed by 4.69, 4.31 and 3.95 mg/I in 7.2, 9.6* and 4.8 g/I concentration of BLA with added 0.4g PSP and 0.2g urea, respectively on 10th day of culture. During the whole culture period the pH range was 6.86 to 8.02.

This experiment was conducted to develop .a suitable Chlorella culture media by varying the amount of banana leaf hsh with added papaya skin powder, and urea.

Preparation of papaya skin powder Papaya skin was dried in an oven at 50°C for 12 hours. It was further dried in the direct sun for seven days. Dried papaya skin was ground into powder with haman dista (Munno Ceramic, Dhaka). Ground papaya skin powder was filtered through 500 mm mesh-sized cloth. Preparation of banana leaf Collected banana leaf was chopped and dried in under the direct sun for seven days. Dried banana leaf was burnt to make ash. Banana ash was filtered through 500 um screen. Preparation of culture media For the preparation of three different banana leaf ash media (BLAM), 7.2, 9.6 and 4.8 g banana leaf ash (BLA) were placed in 1.0 L conical, flask. Then 0.4 g PSP and 0.2 g urea were added in every flask and distilled water was added to make the volume up to 1.0 litre solution. The media were mixed well and covered the mouth • with aluminium foil and sterilized at 12°C for 15 minutes with moist heat in autoclave. Flasks were kept for 24 hrs. for cooling before culture of microalgae. Preparation of control medium Bold basal medium (BBM) was used as control medium. Different composition of inorganic nutrients of BBM, Phang and Chu (1999) is given in Table 1. Mixing, sterilisation and cooling of the prepared BBM was done following the same procedures to culture "media. Culture of C. ellipsoidea 335 Four treatments, three from three various concentrations of banana leaf ash containing 0.4 g/I PSP and 0.2 g urea and one from BBM as control were used to culture Chlorella ellipsoidea. For inoculation C. ellipsoidea was collected from the Live Food Culture Laboratory of Bangladesh Agricultural University (BAU), Mymensingh where the experiment was conducted. Chlorella ellipsoidea were inoculated into culture bottles to produce a culture containing 10% C. ellipsoidea suspension (Optical density 620 nm = 0.25) (Habib, 1998). All the bottles were kept under fluorescent lights (light: dark= 12: 12) in the laboratory. The culture bottles were continuously aerated with plastic tubing. Eight sub-sample were taken at every alternate day from each flasks to observe cell density and chlorophyll-a of C. ellipsoidea and physico-chemical properties of culture media. Temperature, light intensity, dissolved oxygen and pH were determined by respective meters. Nitrite-nitrogen, nitratenitrogen and phosphate-phosphorus were determined by Hach kit (DREU2000). Cell count and estimation of chlorophyll a Chlorella cells were counted using Neubauer ruling haemacytometer and, chlorophyll a was determined by following formula (Clesceri et al., 1989) chlorophyll a (mg/I) = 11.85 (OD 664) -1.54 (OD 647) -0.08 (OD 630) Data analysis Differences among the treatments of C. ellipsoidea in different media were analysed by one way ANOVA (Gomez and Gomez, 1976).

So it was concluded that the growth of C. ellipsoidea was maximum in 7.2 g/I BLAM with added 0.4 g/I PSP and 0.2 g/l urea than other treatment of BLAM. Thus the concentration of 7.2 g/I ash of banana leaf with added PSP and urea is suitable for C. ellipsoidea culture and may be recommended for mass culture.
