Micro Propagation of Papaya (carica Papaya L.) (2018-2019)

Abstract/ Executive Summary:

For the development of a suitable and reproducible protocol for micro propagation of papaya shoot tips of BARI Pepe-1 were cultured on MS medium supplemented with different concentrations of BAP and NAA. The types of responses were different in both BAP and NAA at different concentrations. MS media supplemented with BAP performed better than NAA in all parameters. Among the treatments, BAP at 1.0 mg/l (T₄) and 0.5 mg/l (T₃) were found better for shoot induction of the explants.

Objectives:

To develop a suitable protocol for in vitro clonal multiplication of papaya and make the plant materials available to the farmers for commercial cultivation.

Methodology:

Seeds of BARI Pape-1 (Shahi) were collected from pomology division of HRC, BARI, Gazipur. The seeds were sown in poly bag from November 20, 2018 at one month interval. Shoot tip of two months old seedlings were used as explant. At first the explants were surface sterilized with running tap water for half an hour. Then the explants were sterilized with Jet and Trix for 30 minutes followed by 3-5 times washing with distilled water. The explants were then surface sterilized with Ridomil Gold MZ 68 WG, Tilt and Krosin for another 30 minutes to remove the fungal and bacterial contamination. After that the explants were treated with chlorox solution (60%) with two drops of Tween-20 for 10 minutes followed by 3-5 times washing with double distilled water and again dip in 70% (v/v) ethanol for 2 minutes followed by 5-6 rinses in sterile distilled water inside a laminar airflow cabinet. MS medium containing standard salts and vitamins, 3% sucrose and 0.8% agar was used and p^H of the each medium was adjusted separately to 5.7 prior to addition of agar. Medium was sterilized in autoclave at a temperature of 121⁰C and 15 psi pressure for 20 minutes. The medium was placed in growth room for 3-4 days to check any medial contamination before use for culture of explant. The explants were incubated in a culture room where the temperature was maintained at 25⁰-27⁰C under continuous photo period of 16 h light and 8 h dark.

Achievement/Findings:

From the experiment, it was found that individual effect of NAA has less than BAP on MS media for micro propagation of papaya by shoot tips. Among the treatments, BAP at 0.5 and 1.0 mg/l found better for shoot induction of papaya. The experiment is now on going. So it needs to be continued for the next year to obtain the final result of the experiment.
