The experiment was carried out to investigate the effect of different levels of carbon source into medium on growth and development of PLBs and regenerated plantlets of Dendrobium sonia. The maximum (19.40) number and highest (1.301 g) fresh weight of new PLBs was found at ½ MS medium. The BM was supplemented with different levels of sucrose (0.0, 5.0, 10, 20, 30 and 40 gL?1) as treatments to estimate their effects on growth and development of PLBs and PLB derived plantlets. Among the different levels of sucrose, 30 gL?1  sucrose was the best for PLBs proliferation and plantlets production in vitro. Fresh weight of plantlet was highest (0.051 g) in medium supplemented with 30 gL-1 sucrose. The maximum (27.40) number of PLBs was recorded at 30 gL-1 sucrose. The highest fresh (0.383 g) weight of PLBs per vial was obtained from 30 gL-1 sucrose. The maximum number of shoots (1.2) also found at 30 gL-1 sucrose. The longest shoot (2.918 cm) with the highest (0.057 g) shoot weight were observed in medium supplemented with same concentration level of sucrose. The maximum (3.60) numbers of leaves along with the longest (1.06 cm) leaf were recorded at 30 gL-1 sucrose supplemented in ½ MS Medium. Same level of sucrose was recognized for maximum number of roots (4.98), the longest (1.51 cm) roots and for the highest (0.045 g) fresh weight.

To provide specific informations on growth of PLBs and PLB derived plantlets of Dendrobium sonia in vitro with different levels of sucrose.

The experiment was carried out at the Laboratory of the Department of Biotechnology, Bangladesh Agricultural University, Mymensingh, during the period from February 2009 to May 2010, to investigate the effect of sucrose on in vitro growth and development of Protocorm Like Bodies (PLBs), shoots and roots multiplication of Dendrobium sonia orchid. The PLBs are used as plant materials which were collected from the Tissue Culture Laboratory of Biotechnology Department, Bangladesh Agricultural University, Mymensingh. At first in vitro two months age earlier grown four small pieces PLBs were cultured in Full, Hall and Quarter MS medium. Observations were recorded after two months. Another Six levels of sucrose (0.0, 5.0, 10.0, 20.0, 30.0, 40.0 gL-1) were supplemented into ½ MS medium to estimate their effect on growth, development and proliferation of PLBs and plantlets in vitro. There were no uses of any Plant Growth Regulators (PGRs). Each treatment was replicated five times. The cultured vials were marked for treatments and replication. The culture vials were maintained in a growth chamber and allowed to grow at 25±1OC under 16 hours photoperiod illuminated with florescent tubes of 50 µmol m-2s-2. The data were collected on fresh weight of PLBs, number of PLBs, number of multiple shoots per explants, number of Shoot, length of shoot, weight of shoot, number of leaves per plantlet, length and width of leaves, number and fresh weight of roots, length of roots. The experiment was laid out in Completely Randomized Design (CRD). The analysis of variance was performed and means were compared by Duncan's Multiple Range Test (DMRT) at 5% level of probability for interpretation of results (Gomez and Gomez, 1984). Rate of PLBs proliferation and percent plantlet regeneration were calculated by the following formula-Per cent Plantlet Regeneration= Number of regenerated plantlets from cultured PLB / Number of PLB cultured x 100.

The experiment was conducted to investigate the in vitro PLBs, shoot and root multiplication and development under different concentrations of sucrose. In this study, a series of experiments were conducted with Dendrobium sonia to investigate the effect of sucrose on PLBs multiplication, plantlet regeneration and growth and development of PLBs-derived plantlets. Different concentrations of sucrose on PLBs and shoot and root multiplication and development have been presented. This results also found who showed that ½ MS medium had significant effects on growth and development of PLBs and subsequent plantlets regeneration of Dendrobium orchid. Orchid is the slow growing plant and need comparatively less amount of nutrient for plantlet growth and development. In ½ MS medium the PLBs turn dark green in colour compared to both them. The number of new PLBs regenerated per vial was the maximum (19.40 per vial) in ½ MS medium and the minimum (9.60) was found in ¼ MS medium. Similar scenario is observed incase of Fresh weight of PLBs per vial. The highest (1.301 g) fresh weight was in ½ MS medium followed by MS medium and the lowest (0.443 g) was in quarter MS medium after 60 days of culture. Fresh weight of plantlet was highest (0.051 g) and the lowest (0.025 g) in medium supplemented with 30 gL-1 and 10 gL-1 sucrose, respectively. The 30 gL-1 sucrose is the usual concentration of sucrose used in MS medium, because it gives best performance. The present study proved it once again. The maximum (27.40) numbers of PLBs were observed at 30 gL-1 sucrose whereas the minimum (13.80) PLBs were observed at 5 gL-1 sucrose. This result was supported who reported that 30 gL-1 sucrose produced more PLBs than 30 gL-1 glucose and fructose (combined). The highest (0.383 g) fresh weight of PLBs per vial was obtained from 30 gL-1 sucrose and lowest (0.099 g) was found in 5 gL-1 sucrose. This concentration of sucrose has been advocated. The longest (2.918 cm) and the lowest (0.886 cm) shoot were found at 30 gL-1 and 5 gL-1 sucrose respectively. Further increase of sucrose concentration above 30 gL-1 was found inhibitory for shoot length. The highest (0.057 g) shoot weight was obtained at 30 gL-1 and the lowest (0.030 g) was recorded for 5 gL-1 sucrose. The maximum (3.60) and the minimum (2.00) numbers of leaves were found at 30 gL-1 and 5 gL-1 sucrose, respectively. The longest (1.06 cm) leaf was obtained at 30 gL-1 and the shortest (0.54 cm) was found for 5 gL-1 sucrose. Likewise length, width of leaves was the highest (0.50 cm) with 30 gL-1 sucrose and the lowest (0.32 cm) was found at 5 gL-1 sucrose supplemented in ½ MS medium. After 60 days of culture, the maximum number of roots (4.98) was found at 30 gL-1 sucrose and the minimum (0.98) number was observed at 10 gL-1 sucrose. In medium with 0 (control) and 5 gL-1 sucrose, there was no root in plantlets. Similar trend in length of root was observed. The longest (1.51 cm) and the shortest (0.49 cm) root were obtained from 30 gL-1 and 10 gL-1 sucrose, respectively. The fresh weight of roots per plantlet also varied with different levels of sucrose. The highest (0.045 g) fresh weight was recorded at 30 gL-1 sucrose and the lowest (0.010 g) was for 10 gL-1 sucrose.
