Study of Comparative Regeneration Efficiency of Different Potato Varieties (2014-2015)

Abstract/ Executive Summary:

Internode explants of two BARI released high yielding varieties (BARI Alu-40 and BARI Alu-41) were cultured to compare their regeneration efficiency. Five different concentrations (T₀, T₁, T₂, T₃, & T₄) of Zeatin Riboside in combination with GA₃and IAA were used for regeneration of explants. Among the two varieties, the maximum responding variety was BARI Alu-41 and the best treatment combination was T₃ regarding shoot number, node number and leaf number.

Objectives:

The present study has been undertaken to compare regeneration efficiency of potato varieties.

Methodology:

Tissue culture plantlets of three BARI released recent high yielding varieties, namely BARI Alu-40 (4.45 W), BARI Alu-41 (5.183) and BARI Alu-25 (Asterix) were collected from Tuber Crop Research Centre (TCRC) of BARI. Five plantlets of each variety were obtained. To increase the number of plantlets, those materials were further grown on modified Murashige and Skoog basal medium by node cutting. When sufficient explants were obtained then the experiment was setup. For regeneration, internode explants were cut into small pieces and placed in modified MS media containing different concentrations of Zeatin Riboside, GA₃ and IAA. The P^H of all media was adjusted to 5.8 before autoclaving. The media were autoclaved at 121?C for 20 minutes. All cultures were incubated at 25 ± 1?C and kept in 16/8 hours light/dark photoperiod with a light intensity of 2000 lux under cool white fluorescent tubes.

Achievement/Findings:

Internode explants of three potato varieties were cultured on various Zeatin Riboside (ZR) concentrations along with GA₃and IAA. Reports were made only two varieties. Data of different parameters regarding direct shoot regeneration were obtained six weeks after inoculation.

In the above experiment, result showed that the variety BARI Alu-41 performed better than BARI Alu-40. The T₃ treatment was better for shoot regeneration regarding shoot number, node number and leaf number compared to other treatments.
