This study has been conducted during June 2011 to December 2014. Sword suckers of BARI Kola-3 and BARI Kola-4 were collected from mother Orchard of ARS, Raikhali, Rangamati Hill District. Shoot tips were separated from the suckers and cultured on Murashige and Skoog (MS) medium supplemented with different concentrations (viz. 1, 2, 3, 4, 5 and 6 mg/l BAP) of BAP and 2ip (viz. 1.0, 1.5 and 2.0 mg/l)  to optimize existing protocol for in vitro production of BARI Kola-3 and BARI Kola-4 at Biotech Lab, Gazipur. Among the different concentrations, MS medium supplemented with 4 mg/l BAP was found suitable for in vitro production of multiple shoots in BARI Kola- 3, where 5 mg/l BAP was found suitable in BARI kola-4. Both of the varieties produced well developed roots in ½MS medium supplemented with 0.5 mg/l IBA. Between the varieties, BARI Kola- 4 showed the better performance compare to BARI Kola- 3 for in vitro propagation. After 05 days hardening at room temperature, 100% plantlets were survived when those were transplanted in poly bags and kept 7 days under polyethene tunnel house whereas cent percent (100%) plantlets were died when transplanted plantlets were kept on open conditions at nursery. Plantlets were allowed 1.5-2.0 months at nursery for growth and development before planting at field. No bunchy top virus was found among the plantlets by ELISA testing (using Agdia kit,USA ).

Fifty validation trials were conducted at farmer’s hill at different positions where 36 from tissue cultured plantlets, 10 ratoon crops and 04 from suckers of tissue cultured banana fields. The highest 53.0 t/ha bunch yield was found in BARI Kola-4 at hill valley followed by BARI Kola-3 (47.04 t/ha). Also, BARI Kola-4 produced maximum 39.37 t/ha at hill slope where BARI Kola-3 produced 36.27 t/ha. But in case of hill top, BARI Kola-3 produced maximum 21.19 t/ha where BARI Kola-4 produced 18.25. Validation trial with BARI Kola-3 gave 19.28% more yield than farmer’s practice of BARI Kola-3 and validation trial with BARI Kola-4 gave 28.66% more yield than farmer’s practice with the same variety. In every case, too much low yield was found at hill top having non-marketable fruit size. Tissue cultured BARI Kola-4 produced maximum yield at hill valleys which was 35.53% and 165% higher than hill slope and hill top, respectively. Similarly, BARI Kola-3 also produced 29.67% and 102% higher yield at valley compare to hill slope and hill top, respectively. In case of hill slope, BARI Kola-3 and BARI Kola-4 produced 56.35% and 96.13% more yield than hill top. First ratoon crop gave 10.07% and 13.93% more yield than tissue culture plants in BARI Kola-3 and BARI Kola-4, respectively. Hill valleys have been found suitable for tissue cultured banana production followed by hill base where have minimum irrigation facilities. Due to water scarcity at hilly areas, hill tops and slopes are not suitable for tissue cultured banana cultivation. It should be kept confined on the base of the hills and valleys. The highest 55.89 t/ha bunch yield was found from tissue cultured BARI Kola-4 followed by 49.67 t/ha in BARI Kola-3 at April-May Planting. Tissue cultured BARI Kola-3 and BARI Kola-4 produced 12.75 and 22.37% more yield at April – May planting, respectively compare to September-October planting.

The present project was proposed with aiming at -

  1. To standardize  protocol for in vitro production of BARI Kola-3 and BARI Kola-4
  2. To validate the  performance of tissue cultured banana plantlets in hilly areas

       iii.   To improve knowledge and skill of farmers on tissue cultured banana cultivation in   hilly areas and private entrepreneurs on tissue cultured banana plantlets production 

After successful shoot production, well developed shoots were cultured in MS medium supplemented with IBA for root induction. After 21-28 days, well developed rooted plantlets were kept at room temperature for 5 days and then transferred in poly pot. Transplanted plantlets were hardened under polyethylene tunnel for 7 days and then shifted at nursery conditions. After 45 days, well established plantlets were sent to Agricultural Research Station, Ramgarh, Khagrachari Hill District and Agricultural Research Station, Raikhali , Rangamati Hill District for validation trial establishment.  Before planting, randomly 100 sample plantlets were tested for bunchy top virus using Agdia Bunchy top Virus Testing Kit (USA). Fifty validation trials were established at farmer’s field where 36 from tissue cultured plantlets, 10 ratoon crops and 04 from suckers of tissue cultured banana fields. Only third year findings have been described

      Hill valleys are suitable for tissue cultured banana production followed by hill base where have minimum irrigation facilities. Due to water scarcity at hilly areas, hill tops and slopes are not suitable for tissue cultured banana cultivation due to water scarcity. It should be kept confined on the base of the hills and valleys.

?    March-April plantation has been found suitable for banana cultivation at hilly areas.

?    Market price of BARI kola-3 is higher than the BARI kola-4 and farmers are interested to cultivate BARI kola-3

?    Some farmers produced quick growing vegetables like lalshak, radish, pumpkin, tomato, cotton, bush bean etc. as inter crop in the tissue cultured banana field during the vegetative growth stage. So, farmers have eared Tk. 3000.0 – 4000.0 from each trials (Approx. 0.33 acre of land) from quick growing vegetables but the farmers who cultivated cotton as intercrop earned extra Tk. 9,000/-.
