Validation Trial of Tissue Cultured Plantlets of Jackfruit (2014-2015)

Abstract/ Executive Summary:

Tissue cultured, seeded and grafted saplings of Jackfruit were planted to see the field performance on growth and yield. The highest plant height was found in Tissue cultured plants (5.56 m) followed by seeded (3.92 m) and grafted (3.90 m). The highest base girth (29.3 cm) was also recorded in tissue cultured plants followed by grafted BARI kathal-2 (21.15 cm). Also, the highest number of branch/plant was recorded in tissue cultured plants followed by grafted where the lowest was in the seeded plant. Maximum plant canopy was recorded also in tissue cultured plants followed by grafted and seeded plants both in North-South & East-West direction. There was no male or female flower have been seen in any tissue-cultured, grafted and seeded plants up to the third year. But all of the tissue cultured and grafted plants produced male flowers where one-seeded plant produced fruits in the fourth year.

Objectives:

Thus the investigation was undertaken with an aim to see the field performance of tissue culture saplings of Jackfruit compared to grafted and seeded ones.

Methodology:

The regenerated rooted plantlets with 5-6 leaf stage were transferred from the culture room and kept at room temperature (30⁰C) for 5-7 days. The plantlets were then removed from the culture tubes and all the adhering media were carefully washed out so that, the root damage was the least. Washed plantlets were planted into small polyethylene bags containing sterile soil, sand and decomposed cow dung at the ratio of 1: 1: 1. Subsequently, the healthy plants were transplanted in a 9 x 12-inch earthen pots containing soil mixture at the nursery for three months. Well established fifteen saplings were transplanted along with grafted and seeded ones at Biotech Field, Gazipur; Hill Agricultural Research Station, Khagrachari Hill Districts; Hill Agricultural Research Station, Ramgarh, Khagrachari Hill Districts; Regional Agricultural Research Stations, Hathazari, Chittagong & Akbarpur, Sylhet and Oodaleah Tea Estate, Nagirhat, Chittagong on May 2011. Saplings were transplanted at a single-line design following plant to plant distance of 10.0 m. A single plant was considered as one replication. Data on Plant height, base girth and number of branches/plant and plant canopy were recorded.

Achievement/Findings:

Cent percent (100%) transplanted saplings were survived at field conditions. Data on different parameters have been presented. Among the three treatments, the highest plant height was found in Tissue cultured plants (5.56 m) followed by seeded (3.92m) and grafted (3.90 m). The highest base girth (29.3 cm) was recorded in tissue cultured plants followed by grafted (21.15 cm). Also, the highest number of branch/plant (9.4) was recorded in tissue cultured plants followed by grafted plants (7.33) where the minimum was in the seeded plant (5.89). Plant canopy was also found higher in tissue cultured plants (N-S = 2.79 m, E-W = 2.87 m) compare to seeded and grafted ones, respectively (N-S = 1.80 m, E-W = 1.73 m and N-S = 2.14 m, E-W = 2.01m (Table 1). All of the tissue-cultured and grafted plants produced male flower but one-seeded plant produced fruits in the fourth year. Significant results have not yet been generated. So, the investigation should be continued for the next years.
