Generation Advancement of Rest Bt Brinjal Lines (2014-2015)

Abstract/ Executive Summary:

A field trial on generation advancement of five Bt Brinjal lines was conducted at the confined field of Biotechnology Research Farm, BARI Gazipur during 2014-15. Five Bt Brinjal lines viz. Bt Dohazari, Bt Shinghnath, Bt Khatkhatia, Bt Islampuri and Bt Chega were tested along with its counter non-Bt varieties. Considering the distinctiveness, uniformity and stability of the lines, Bt Dohazari may be proposed to release as GE crop. A slight variation was observed in Bt Shinghnath and Bt Khatkhatia, under close observation, it may be released as GE crops as soon as possible. More purification is needed in the case of Bt Islampuri and Bt Chega.

Objectives:

The breeder must ensure that a level of uniformity and stability exists throughout multiplication and commercialization. After the required backcrosses, selfing and multilocation trials, the programme on generation advancement of the rest of the lines was conducted at the confined field of the Biotechnology Division.

Methodology:

Seedlings of BC₃F₆of Bt brinjal lines were grown at the contained greenhouse of the Biotechnology division. Considering the Bt strip test, the 40-45-day seedlings having the positive result were transplanted at the confined field on November 24, 2014. The unit plot size was 56 m² and it was replicated in four. The spacing was 1m x 1m. The counterparts of the Bt brinjals (i.e. non-Bt) were also planted at the different block to compare its homozygosity. To ensure genetic purity, the Bt plants were covered by the fine mosquito net before the flowering stage. Frequent visits were made by the scientists. At the fruiting stage, off-types were discarded. Other intercultural operations such as weeding, mulching, fertilizer application, irrigation, sticking etc. were performed as and when necessary. Admire (to control whiteflies, jassid) and Vertimec (to control mies) were also applied as and when necessary. At the final stage, the number of plants of Bt Dohazari, Bt Khatkhatia, Bt Shingnath, Bt Islampuri and Bt Chega was 200, 179, 203, 181 and 196, respectively.

Achievement/Findings:

There was no incidence BFSB in the brinjal field. Distinctiveness, uniformity, stability and homozygosity etc. were considered for the generation advancement.

Bt Dohazari : The plants were found as dwarf intermediate, light green stem colour, solitary and cluster bearing habit, oval-shaped fruit and green with white stripe fruit colour.

Bt Shingnath : The plants were found as upright growth habit, purple-green stem colour, solitary and cluster bearing habit, snake or sickle-shaped fruit shape. Comparing the non-Bt, slight variation was found in the fruit colour.

Bt Khatkhatia: The plants were found as upright growth habit, purple-green stem colour, solitary and cluster bearing habit, cylindrical fruit shape. Comparing the non-Bt, slight variation was found in the fruit colour.

Bt Islampuri: The plants were found as an intermediate habit, dark green stem colour, solitary bearing habit, large fruit shape. Comparing the non-Bt, variation was found in the fruit colour.

Bt Chega: The plants were found as intermediate growth habit, light green stem colour, solitary bearing habit, oval fruit shape. Comparing the non-Bt, variation was found in the fruit colour.

Considering the field performance, Bt Dohazari may be proposed as a genetically engineered variety and Bt Shinghnath and Bt Khatkhatia should be under close observation to release as GE variety very soon. More purification is needed for Bt Islampuri and Bt Chega.
