Standardization of Protocol for Advanced Lines of Strawberry and Their Large Scale Multiplication (2015-2016)

Abstract/ Executive Summary:

Shoot tips of strawberry lines SB003 and SB005 were cultured on MS medium supplemented with a combination of BAP, Kn and GA₃ along with a control. Shoot initiation was started within 6-7 days after culture. Five different treatments (T₀, T₁, T₂, T₃ and T₄) were used to shoot the multiplication of explants. Percentage of explant-producing shoots were best in T₄ treatment for both lines. In the case of SB003 T₃ treatment and for SB005 T₂ and T₃ were best regarding shoot number, plant light and leaf number. ½ MS medium without hormone was used for root induction. Rooting performance was good in ½ MS medium for both lines. A small number of plantlets were transferred in a polybag containing soil and cowdung and kept in the net house condition.

Objectives:

Therefore, the present experiment is important to develop a protocol for mass propagation of introduced lines of strawberry

Methodology:

Strawberry lines (SB003, SB005) collected from the fruits field of HRC, BARI were used as materials. Small shoot tip came out from runner were carefully removed by scalpel from Strawberry plants showing no virus symptoms. The explants were prepared by removing the outer layers of tissue from the shoot tips by the scalpel. The light greenish 2-3 cm shoot tips were surface sterilized with Ridomil Gold MZ 68WG for 5 min. to remove the fungal contamination. Then shoot tips were sterilized with Jet and Trix under running tap water for 5-6 min followed by 3-4 times washing with distilled water. After then shoot tips were sterilized with Clorox solution (60 ml Clorox: 40 ml distilled water) with 2 drops of Tween-20 for 3min. followed by 3-4 times washing with sterile distilled water inside the laminar airflow cabinet. The dead tissue of sterilized explants was cut off to leave 1-2 cm portion containing an intact apex. The prepared explants were placed in MS media (Murashige and Skoog, 1962) with different concentrations and combinations of BAP, Kn and GA₃. All cultures were incubated at 25 ± 1⁰C with a 16 h photoperiod (2000 lux) provided by cool white fluorescent tubes. The pH of the medium was adjusted to 5.8.

Shoot tip of two lines SB003 and SB005 of strawberry were cultured on MS medium supplemented with five different combinations of BAP, Kn and GA₃ with control. showed that the percentage of explant-producing shoots varied among the treatments. The maximum responding explant (40%) was obtained from T₄ treatment in line SB003 and T₃ and T₄(40%) for line SB005. The minimum responding explants (13% and 15%) from T₁ treatment in both lines. T₀ treatment did not produce any shoots in both lines. From SB003 lines the highest number of shoots (7.15) per explant was observed in treatment T_3,2^nd highest was T₂ and the lowest was in T₁ treatment. The highest shoot length (3.65 cm) and a maximum number of leaves (5.3) were also in treatment T₃ and 2^nd highest (3.00), (4.90) was T₂ and the lowest (2.65, 3.30) in T₁ respectively. From SB005 line the highest number of shoots per explant (6.2 and 6.1) was found in both treatment T₂ and T₃ and the lowest was in treatment T_1.The highest shoot length (3.17, 3.00) and maximum leaves number (5.35, 5.30) were found in treatment T₂ and T₃ and lowest in T₄ treatment. For rooting ½ MS medium without hormone was used and performed better for root induction in two lines regarding root numbers and root length. The small number of plantlets were transferred in polybag containing soil and cowdung and kept in the net house condition.
