Internode explants of three BARI released high-yielding potato varieties (BARI Alu-40, BARI Alu-41, BARI Alu-25) and one exotic (Maris Piper) varieties were cultured to compare their regeneration efficiency. Five different concentrations (T0, T1, T2, T3, & T4) of Zeatin Riboside in combination with GA3 and IAA were used for regeneration of explants. Among the four varieties, the maximum responding variety was BARI Alu-41 and the best treatment combination was T2 regarding percent shoot response, shoot height and leaf number.

The present study has been undertaken to compare the regeneration efficiency of potato varieties.

 

One exotic (Maris Piper) and three BARI released high-yielding potato varieties viz. BARI Alu-40 (4.45 W), BARI Alu-41 (5.183) and BARI Alu-25 (Asterix) were used for this study. Tissue culture plantlets of those three varieties were collected from Tuber Crop Research Centre (TCRC) of BARI. Five plantlets of each variety were obtained. To increase the number of plantlets, those materials were further grown on modified Murashige and Skoog basal medium by node cutting. When sufficient explants were obtained then the experiment was set up. For regeneration, internode explants were cut into small pieces and placed in modified MS media containing different concentrations of Zeatin Riboside, GA3 and IAA. The PH of all media was adjusted to 5.8 before autoclaving. The media were autoclaved at 121?C for 20 minutes. All cultures were incubated at 25 ± 1?C and kept in 16/8 hours light/dark photoperiod with a light intensity of 2000 lux under cool white fluorescent tubes.

Internode explants of four potato varieties were cultured on various Zeatin Riboside (ZR) concentrations along with GA3 and IAA. Data of different parameters regarding direct shoot regeneration were obtained six weeks after inoculation. The percentage of explant-producing shoots varied among the four varieties. The maximum responding explant (88%) was obtained from the variety BARI Alu-41 followed by Asterix (78%) and the lowest from Maris Piper (75%). Percent of explant-producing shoots varied in response to different concentrations of ZR. The maximum explant-producing shoot (99%) was recorded at T2 followed by T1 (97%). When considering each hormonal combination individually with each variety, variation was observed. In BARI Alu-40 variety, the maximum explant-producing shoot (100%) was noticed in T2 followed by T3 (94%). In BARI Alu-41, the maximum explants producing shoot (100%) in T1 to T4 . In Asterix, the maximum explants producing shoot (100%) in T1 & T2 treatment. On the other hand, Maris Piper showed maximum explants producing shoot (100%) in T1 & T4 treatment. The minimum response (13%) was observed in control treatment from the variety BARI Alu-40 but in Maris Piper, control treatment did not produce any shoot. The highest number of shoots per explant (3.13) was recorded from BARI Alu-41 and the lowest (1.52) was recorded from Asterix. Different levels of ZR influenced the number of shoots per explant. The highest number of shoots per explant (3.07) was recorded in T3 and the lowest (0.16) was recorded from the control treatment (T0). Distinct variations were found among variety toward various ZR concentrations. The maximum number of shoots per explant (7.51) was recorded from the variety BARI Alu-41 with T3 followed by BARI Alu-40 at T2 (4.63). No shoots were observed from the control treatment of Maris Piper. Results revealed that variety BARI Alu-41 has high regeneration potential than the other three varieties. Shoot height varied among the varieties. The maximum height (3.10 cm) was recorded from BARI Alu-41 followed by Maris Piper (2.94 cm). On the other hand, the lowest (1.92 cm) was observed from BARI Alu-40. Shoot height varied at different concentrations of ZR. The maximum height (3.61 cm) was recorded in T2 followed by T1 (3.07cm). The minimum height (0.59cm) was recorded in the control treatment (T0). In response to various concentrations of ZR, the tested varieties showed a varied response. The maximum shoot height (4.59 cm) was observed in T1 at Maris Piper followed by T2 (4.18 cm) of Asterix variety. Node numbers varied among the varieties. The maximum node number (3.45) was obtained from Maris Piper followed by BARI Alu-41 (3.05). The minimum node number (2.34) was obtained from Asterix. Node numbers also varied at different levels of ZR. The maximum node number (3.70) was recorded in T1 followed by T2 (3.40) and the minimum (0.77) was observed in the control treatment (T0) (1.13). In response to various concentrations of ZR, the tested varieties showed different responses. The maximum node number (5.82) was observed in T4 at Maris Piper followed by T1 (5.25) at the same variety and T2 (4.94) at BARI Alu-40.  Variation was found among the varieties in leaf numbers. The maximum leaf number (4.27) was obtained from BARI Alu-41 followed by Maris Piper (4.26). The minimum leaf number (2.34) was obtained from Asterix. Leaf number also varied at different levels of ZR. The maximum leaf number (4.82) was recorded in T2 followed by T1 (4.65) and the minimum (0.97) was observed in the control treatment (T0). In response to various concentrations of ZR, the tested varieties showed a different response. The maximum leaf number (7.07) was observed in T4 at Maris Piper followed by T1 (6.26) at the same variety and T2 (6.13) at BARI Alu-40. In vitro grown shoots were used for root induction. For rooting, the full strength of PROP medium was used. In this experiment, no rooting hormones were used. Only basal PROP medium was enough for root induction. Similar results were obtained by Chin et al. (1990) and Khan et al. (1994) in their experiments where they used half-strength MS only for root induction in chrysanthemum
