Molecular Characterization of Natural Edible Mushrooms in Bangladesh (2016--2017)

Abstract/ Executive Summary:

Ten natural edible mushrooms were collected from Khagrachari and Moulvibazar districts of Bangladesh during April-June, 2017. Only 4 mushroom samples (sample 1, 6, 7 and 9) could be identified up to the genus level by morphological observation and all of those belong to the genus Pleurotus. In case of tissue culture, among 10 samples only one sample (i.e S-6) could be grown on PDA medium and others did not survive. Mycelium of the culturable sample was white in colour, size of the colony on the 8^th day of culture was 3.1cm and the density of colony was deep. In the case of molecular characterization, DNA extraction was completed for all the samples collected.

Objectives:

The present investigation has been undertaken to identify and characterize the natural edible mushrooms of Bangladesh at a molecular level.

Methodology:

Collections of different natural edible mushroom were done in April-June, 2017. The collection sites were forests, the local market and nearby villages of Khagrachari and Moulvibazar districts of Bangladesh. The collected mushrooms were packed into butter paper bags sand stored in the refrigerator until processing. Morphological characteristics such as the shape of pileus and stipe, surface colour, the apex of the pileus, margin pattern, stipe attachment to pileus, surface pattern, annulas/ring of stipe, gill attachment and gill arrangement were observed visually. Using tissue culture techniques, the mushrooms were aseptically inoculated in Potato Dextrose Agar (potato 200g/l, dextrose 20g/l, agar 20 gl/1,) media and incubated at 25±2°C with a 16 h photoperiod (2000 lux) provided by fluorescent tubes to develop mycelium. For mother culture preparation, wheat grains were thoroughly washed, soaked (20-30 min) and boiled (20-25 min) in water (1.75 litre/kg grain). Excess water from the boiled grains was removed by spreading on the sieve and air drying for few hours. Then Calcium carbonate (5g/kg dry weight basis) was mixed with the grains to adjust the pH around 7.0. Glass bottles were filled up to 2/3 volume with 300 g prepared substrate and plugged with non-absorbent cotton. The plugs were covered with brown paper. These bottles were then autoclaved at 121⁰C at 22 lb psi pressure and kept in the room for 24 hours for cooling. A piece of mycelium from pure culture was aseptically transferred to these bottles and incubated under similar culture conditions as used for pure culture preparation. Genomic DNA was extracted following standard procedures and preserved at -20º C for further study.
